miR-125a-5p在视神经炎患者PBMC中的表达及其生物信息学分析 被引量:1

摘要 目的:探讨miR-125a-5p、miR-145-3p和miR-145-5p在视神经炎(ON)患者外周血单个核细胞(PBMC)中的表达水平,并采用生物信息学方法分析差异表达的microRNA在ON发病过程中的分子调控网络。方法:收集ON患者5例(ON组)及同期体检健康者5例(对照组),应用实时荧光定量聚合酶链反应(qPCR)法检测PBMC中miR-125a-5p、miR-145-3p和miR-145-5p的表达水平。应用miRNAbase数据库分析miR-125a-5p在不同物种间的保守性。应用TargetScan数据库、PicTar数据库、miRanda数据库及miRDB数据库预测hsa-miR-125a-5p交集靶基因;应用DAVID数据库对hsa-miR-125a-5p靶基因进行GO功能富集分析;应用KOBAS数据库对hsa-miR-125a-5p靶基因进行KEGG信号通路富集分析;应用STRING数据库对hsa-miR-125a-5p靶基因进行蛋白-蛋白互作(PPI)网络分析。结果:ON组PBMC中miR-125a-5p表达水平(1.086×10^-3±0.692×10^-3)显著低于对照组(4.468×10^-3±2.322×10^-3)(P<0.05),两组miR-145-3p和miR-145-5p表达比较,均未见明显差异(P>0.05);miR-125a-5p成熟序列在不同物种间具有高度保守性;hsa-miR-125a-5p潜在交集靶基因有78个;GO富集结果显示,hsa-miR-125a-5p靶基因功能主要富集于nucleus、nucleoplasm、membrane等14个生物学过程;KEGG通路结果显示,hsa-miR-125a-5p靶基因主要富集于MicroRNAs in cancer、PI3K-Akt signaling pathway等17条信号通路;PPI网络结果显示,hsa-miR-125a-5p靶基因主要作用于DICER1等72种蛋白。结论:ON患者PBMC中hsa-miR-125a-5p表达水平升高,其可能在多条信号通路中发挥作用,参与ON的发病过程。 Objective: To explore the expression of miR-125 a-5 p,miR-145-3 p,and miR-145-5 p in peripheral blood mononuclear cells(PBMC)of patients with optic neuritis(ON),and to analyze the molecular regulatory network of differentially expressed microRNA in the pathogenesis of ON by bioinformatics method.Methods: The expressions of miR-125 a-5 p,miR-145-3 p and miR-145-5 p in PBMC were detected by real-time fluorescence quantitative polymerase chain reaction(qPCR)in 5 patients with ON(ON group)and 5 healthy subjects(control group).MiRNAbase database was used to analyze the conservation of miR-125 a-5 p among different species.TargetScan database,PicTar database,miRanda database,and miRDB database were used to predict the intersection target genes of hsa-miR-125 a-5 p.DAVID database was used to analyze GO functional enrichment of hsa-miR-125 a-5 p target genes.KOBAS database was used to analyze KEGG signal pathway enrichment of hsa-miR-125 a-5 p target genes.STRING database was used to analyze protein-protein interaction(PPI)network of hsa-miR-125 a-5 p target genes.Results: The expression level of miR-125 a-5 p in PBMC of ON group(1.086×10^-3±0.692×10^-3)was significantly lower than that of control group(4.468×10^-3±2.322×10^-3)(P<0.05).There was no significant difference in the expression of miR-145-3 p and miR-145-5 p between the two groups(P>0.05).The mature sequence of miR-125 a-5 p was highly conserved among different species,and there were 78 potential target genes of hsa-miR-125 a-5 p.The results of GO enrichment showed that the functions of hsa-miR-125 a-5 p target genes were mainly enriched in 14 biological processes(BP)such as nucleus,nucleoplasm,membrane,and so on.The results of KEGG signal pathway showed that hsa-miR-125 a-5 p target genes were mainly enriched in 17 signal pathways such as MicroRNAs in cancer and PI3 K-Akt signaling pathway.The results of PPI network showed that hsa-miR-125 a-5 p target genes mainly acted on 72 proteins such as DICER1.Conclusion: The expression of hsa-miR-125 a-5 p is increased in PBMC of ON patients,which may play a role in multiple signal pathways and participate in the pathogenesis of ON.
