Determination of Seven Constituents in Weiyan Capsules

目的:建立胃炎胶囊中7种成分含量的测定方法。方法:采用HPLC法,色谱柱为Agilent ZORBAX SB-C18(250 mm×4.6 mm,5μm);流动相:5%四氢呋喃乙腈溶液-0.1%磷酸水溶液为流动相,梯度洗脱;流速:1.0 ml·min^-1;检测波长:260 nm;柱温:30℃;进样量:10μl;洗脱时间:80 min。结果:三七皂苷R1在0.257~5.140μg(r=0.999 9)、人参皂苷Rg1在0.599~11.980μg(r=0.999 9)、芦荟大黄素在0.036~0.716μg (r=0.999 9)、大黄酸在0.049~0.972μg (r=0.999 8)、大黄素在0.036~0.716μg(r=0.999 9)、大黄酚在0.044~0.872μg (r=0.999 5)、大黄素甲醚在0.022~0.440μg (r=0.999 7)范围内线性关系良好。上述成分的平均回收率分别为98.45%,96.69%,96.87%,99.25%,98.85%,97.41%,96.39%,对应的RSD分别为1.23%,1.84%,1.77%,1.02%,1.10%,1.43%,1.68%(n=6)。结论:所建方法准确、灵敏、简便,可用于胃炎胶囊的质量控制。

Objective: To establish the determination method for seven constituents in Weiyan capsules. Methods: The HPLC column was Agilent ZORBAX SB-C18( 250 mm×4.6 mm,5 μm). The mobile phase was 5% tetrahydrofuran acetonitrile-0.1% phosphoric acid with gradient elution at the flow rate of 1.0 ml·min^-1;the detection wavelength was at 260 nm;the column temperature was30℃;the injection volume was 10 μl;the elution time was 80 min. Results: The linear range of notogenoside R1,gensenoside Rg1,aloe-emodi,thein,emodin,chrysophanol and physcion was 0.257-5.140 μg( r = 0.999 9),0.59-11.980 μg( r = 0.999 9),0.036-0.716 μg( r = 0.999 9),0.049-0.972 μg( r = 0.999 8),0.036-0.716 μg( r = 0.999 9),0.044-0.872 μg( r = 0.999 5) and 0.022-0.440 μg( r = 0.999 7),the corresponding recovery was 98.45%,96.69%,96.87%,99.25%,98.85%,97.41% and 96.39%,and the corresponding RSD was 1.23%,1.84%,1.77%,1.02%,1.10%,1.43% and 1.68%( n = 6),respectively. Conclusion: The method is accurate,sensitive and simple,which can be used for the quality control of Weiyan capsules.
