猫爪草纳米乳喷雾剂的制备及对慢性咽炎大鼠模型的影响研究

Preparation of Ranunculi Ternati Radix Nano Emulsion Spray and Its Effect on Rat Model of Chronic Pharyngitis

目的:制备猫爪草纳米乳喷雾剂并初步考察其对慢性咽炎大鼠模型的影响。方法:通过溶解度测定,选择溶解猫爪草提取物能力较强的油相、乳化剂和助乳化剂,绘制伪三元相图比较纳米乳区域的大小以确定辅料种类及Km值;考察猫爪草纳米乳喷雾剂对大鼠慢性咽炎模型病理组织的影响,对局部病理检查结果进行半定量分析。结果:以蓖麻油聚氧乙烯醚(EL-40)-甘油-肉豆蔻酸异丙酯(IPM)-水(质量比为21∶7∶10∶61)制得的猫爪草纳米乳性状良好,平均粒径为(84±3. 12) nm,多分散指数(PDI)为0. 106,体系稳定。大鼠慢性咽炎模型病理检查结果表明,阳性药对照组、猫爪草纳米乳喷雾剂(高、中)剂量组的大鼠咽部病理学评分显著低于模型组。结论:猫爪草纳米乳喷雾剂制备简单,纳米乳粒径小,对慢性咽炎具有一定的疗效。

Objective To product the Ranunculi Ternati Radix nano emulsion spray and determine its effect on the rat model of chronic pharyngitis. Methods The oil phase,emulsifiers and co-emulsifiers were selected by solubility test and phase diagram studies.The effects of the Ranunculi Ternati Radix nano emulsion spray on the pathological tissues of rats with chronic pharyngitis were investigated,and the results of local pathological examination were semi-quantitatively analyzed. Result The Ranunculi Ternati Radix nano emulsion was prepared with Polyoxyethylene castor oil EL-40,Glycerol,Isopropyl Myristate and Water( W-W = 21∶ 7∶ 10∶ 61) and showed good physical properties. The average particle size of the Ranunculi Ternati Radix nano emulsion was( 84 ± 3. 12) nm. The poly-dispersity index( PDI) was 0. 106,respectively,which proved that theRanunculi Ternati Radix nano emulsion formed a stable system. The pathological examination results of the rat chronic pharyngitis model showed that compared with the model group,the degree of mucosal epithelial hyperplasia and the degree of inflammatory cell infiltration in the pharyngeal tissue of the positive drug control group and the cat claw grass nanoemulsion spray( high and medium) dose group were significantly reduced. Conclusion The Ranunculi Ternati Radix nano emulsion is not only easy to prepare has small particle size,but also has a certain effect on chronic pharyngitis.
