慢性肾病大鼠心功能损伤情况及其与内质网应激的关系

Condition of cardiac dysfunction and its relationship with endoplasmic reticulum stress in rats with chronic kidney disease

摘要 目的探讨慢性肾病大鼠心功能损伤情况及其与内质网应激的关系。方法将16只SD大鼠随机分为对照组和慢性肾病组,每组8只。慢性肾病组采用5/6肾切除法制备慢性肾病大鼠模型,对照组只撕脱肾包膜,不切除肾脏。肾切除术12周后,比较两组大鼠肾功能、心功能指标以及心肌组织中内质网相关蛋白表达水平。结果肾切除术12周后,慢性肾病组大鼠血尿素氮和肌酐水平、心脏重量指数、收缩压和左室舒张末压均高于对照组(均P<0.05);慢性肾病组大鼠心肌组织葡萄糖调节蛋白94、转录激活因子(ATF)4、ATF6和CCAAT增强子结合蛋白同源蛋白的表达水平均高于对照组(均P<0.05)。结论慢性肾病大鼠存在心功能损伤,内质网应激及其诱导的细胞凋亡可能参与该过程。 Objective To explore the condition of cardiac dysfunctition and its relationship with endoplasmic reticulum stress in rats with chronic kidney disease(CKD).Methods Sixteen SD rats were randomly divided into control group and CKD group,with eight rats in each group.The CKD group used 5/6 nephrectomy to develop a rat model of CKD,while the control group received renal capsule evulsion alone without nephrectomy.Twelve weeks after nephrectomy,renal function,cardiac function indicators,and the expression levels of endoplasmic reticulum-related proteins in myocardial tissue were compared between the two groups.Results Twelve weeks after nephrectomy,the levels of blood urea nitrogen and creatinine,cardiac mass index,systolic blood pressure,and left ventricular end-diastolic pressure in the CKD group were higher than those in the control group(all P<0.05);the CKD group had higher expression levels of glucose-regulated protein 94,activating transcription factor(ATF)4,ATF6 and CCAAT enhancer-binding protein-homologous protein compared to the control group(all P<0.05).Conclusion Cardiac dysfunction exists in rats with CKD,and endoplasmic reticulum stress as well as cell apoptosis induced by endoplasmic reticulum stress may be involved in this process.
