无症状生殖道感染的男性不育患者γH2AX、白细胞亚群与精子DNA损伤关系研究 被引量:2

摘要 目的分析无症状生殖道感染的男性不育患者γH2AX、白细胞亚群与精子DNA损伤的关系。方法选取2016年8月至2018年8月银川市妇幼保健院泌尿外科门诊诊治的94例无症状生殖道感染男性不育患者作为研究对象。将这94例患者作为观察组,另选取同时期进行健康体检的94例有生育能力的健康男性作为对照组。采用免疫细胞法、流式细胞术检测精液中CD45^+白细胞、巨噬细胞系CD14^+抗原、活化巨噬细胞细胞HLA-DR^+细胞浓度及γH2AX含量,之后比较两组受试者精液中γH2AX、白细胞亚群水平与8-OHDG表达情况。结果观察组患者的精液γH2AX百分含量,CD45^+、CD14^+、HLA-DR^+白细胞亚群水平及8-OHDG精子百分率显著高于对照组受试者精液,差异具有统计学意义(P<0.05)。精液γH2AX百分含量与8-OHDG精子百分率的呈正相关,差异具有统计学意义(r=0.295,P<0.05);CD45^+、CD14^+、HLA-DR^+等白细胞亚群与8-OHDG精子百分率呈正相关,差异具有统计学意义(r=0.376,r=0.191,r=0.252,P<0.05)。结论无症状生殖道感染的男性不育患者精液γH2AX百分含量、CD45^+、CD14^+、HLA-DR^+白细胞亚群水平显著高于健康男性,且精子DNA损伤与γH2AX含量、不同白细胞亚群浓度呈正相关。 Objective To analyze the relationship betweenγH2 AX,leukocyte subsets and sperm DNA damage in infertile patients with asymptomatic reproductive tract infection.Methods A total of 94 male infertile men with asymptomatic genital tract infections(observation group)who were treated in the urology clinic of our hospital from August 2016 to August 2018 and 94 healthy men for health examination(control group)at the same time were selected.The concentration of CD45^+leukocytes in semen,the macrophage cell line CD14^+antigen,activated macrophage cell HLA-DR^+cell concentration andγH2 AX content were detected by immunocytochemistry and flow cytometry.The levels ofγH2 AX,leukocyte subsets and 8-hydroxydeoxyguanosine(8-OHDG)in semen were compared between the two groups.Results The percentage of semenγH2 AX,CD45^+,CD14^+,HLA-DR^+leukocyte subsets and 8-OHDG sperm percentage in the observation group were significantly higher than those in the control group,with statistically significant differences(P<0.05).The percentage of semenγH2 AX was positively correlated with the percentage of 8-OHDG sperm(r=0.755,P<0.05).The percentage of leukocyte subsets such as CD45^+,CD14^+and HLA-DR^+was positively correlated with the percentage of 8-OHDG sperm(r=0.633,r=0.458,r=0.475,P<0.05).Conclusions The percentages of semenγH2 AX,CD45^+,CD14^+,HLA-DR^+leukocyte subsets in male infertile patients with asymptomatic genital tract infection are significantly higher than those in healthy men,and sperm DNA damage is positively correlated withγH2 AX content and different leukocyte subsets concentration.
