摘要
目的探讨广西地区565例肝细胞性肝癌(HCC)患者和561例健康对照者CD44单核苷酸多态性(SNPs)与HCC易感性及临床病理特征的关系。方法采用SNaPshot方法对CD44 rs187115进行基因分型,非条件Logistic回归分析法对rs187115位点等位基因和基因型频率与HCC的发病风险关系进行分析。结果AG杂合子和GG纯合子基因型,以及G载体状态和G等位基因与发生HCC风险增加有关;与携带至少一个等位基因(AG、AG+GG和G等位基因)的个体相比,携带GG纯合子的个体发生HCC的风险更高。此外,在CD44 rs187115多态性和临床特征亚组分析中发现CD44 rs187115的基因型在转移亚组,肝硬化亚组和ALT阳性和阴性亚组中存在显著差异。结论CD44 rs187115基因多态性与我国南方地区肝癌发病有一定相关性,CD44 rs187115可作为预测肝细胞癌发生发展的标志物,但仍需要更大规模的样本研究。
Objective To explore the correlation between CD44 Single nucleotide polymorphism(SNPs)and HCC susceptibility and clinicopathological features in 565 patients with Hepatocellular carcinoma(HCC)and 561 healthy controls in Guangxi.Methods CD44 rs187115 was genotyped through SNaPshot method,and the relationship between allele and genotype frequency of rs187115 locus and the risk of HCC was analyzed through unconditional Logistic regression analysis.Results AG heterozygote and GG homozygote genotypes,G vector status and G allele were associated with increased risk of HCC.Compared with individuals carrying at least one allele(AG,AG+GG and G allele),individuals carrying GG homozygotes have a higher risk of HCC.In addition,in the subgroup analysis of CD44 rs187115 polymorphism and clinical characteristics,it was found that the genotype of CD44 rs187115 varied greatly among the metastasis subgroup,cirrhosis subgroup and ALT positive and negative subgroup.Conclusion CD44 rs187115 gene polymorphism has a certain correlation with the incidence of hepatocellular carcinoma in southern China,and CD44 rs187115 can be used as a marker to predict the occurrence and development of HCC,but it still needs more extensive sample research.
