宫颈癌组织中同源形成素样蛋白2、HERC4表达量与癌细胞浸润性生长的关系研究 被引量:1

Study on the relationship between homologous formin-like protein 2 and HERC4 expression quantity in cervical cancer tissue and infiltrative growth of cancer cells

摘要 目的探究宫颈癌癌细胞浸润性生长与组织中同源形成素样蛋白2(FMNL2)、E3泛素连接酶HERC4(HERC4)表达量的关系。方法对我院2017年2月~2018年2月收治的80例宫颈癌手术患者展开研究,术后收集入组患者的宫颈癌组织和癌旁组织,测定FMNL2、HERC4含量,比较不同含量的FMNL2、HERC4组织抑癌基因、增殖基因、侵袭基因的荧光定量PCR表达量。结果在FMNL2、HERC4表达上,宫颈癌组织显著比癌旁组织高(P<0.05),宫颈癌组织在外泌体(Exosomes)、E盒结合锌指蛋白1(ZEB1)、波形蛋白(Vimentin)、畸胎瘤衍生生长因子(Cripto-1)、锌指转录因子Snail(Snail)上均比癌旁组织高(P<0.05),高FMNL2在Exosomes、ZEB1、Vimentin、Cripto-1、Snail上均比低FMNL2高(P<0.05),宫颈癌组织在干细胞蛋白Piwil2(Piwil2)、海马B淋巴细胞瘤-2(BCL2)、X-linked细胞凋亡抑制蛋白(XIA)、Stomatin样蛋白2(STOML2)、精胺氧化酶(Smo)上均比癌旁组织高(P<0.05),高HERC4在Piwil2、BCL2、XIA、STOML2、Smo上均比低HERC4高(P<0.05)。结论宫颈癌组织中FMNL2、HERC4含量较高,FMNL2、HERC4含量会影响癌细胞浸润性生长。 Objective To explore the relationship between infiltrative growth of cervical cancer cells and homologous formin-like protein(FMNL2)in the tissue and E3 ubiquitin ligase HERC4(HERC4)expression quantity.Methods A study was conducted on 80 patients with cervical cancer who received surgery at our hospital from February 2017 to February 2018.The cervical cancer tissue and paracarcinoma tissue were collected from patients after operation to determine FMNL2 and HERC4 contents.The fluorescent quantitation PCR expression quantities of cancer suppressor gene,hyperplasia gene and invasion gene of FMNL2,HERC4 tissue at varied contents were compared.Results The FMNL2 and HERC4 expression of cervical cancer tissue were significantly higher than paracarcinoma tissue(P<0.05),and Exosomes,E-box binding zinc finger protein(ZEB1),Vimentin,teratocarcinoma-derived growth factor(Cripto-1)and zinc finger transcription factor Snail(Snail)were higher than those of paracarcinoma tissue(P<0.05),while high FMNL2 expression in Exosomes,ZEB1,Vimentin,Cripto-1 and Snail were all higher than low FMNL2 expression(P<0.05).The stem cell protein Piwil2(Piwil2),B-cell lymphoma-2 in hippocampus(BCL2),X-linked inhibitor of apoptosis protein(XIA),Stomatin-like protein 2(STOML2)and spermine oxidase(Smo)in cervical cancer tissue were higher than those in paracarcinoma tissue(P<0.05),and high HERC4 expression in Piwil2,BCL2,XIA,STOML2 and Smo were higher than low HCER4 expression(P<0.05).Conclusion The contents of FMNL2 and HERC4 in cervical cancer tissue were high,which would affect the infiltrative growth of cancer cells.
