莫沙必利用于小鼠试验最佳剂量筛选

摘要 为了筛选用于小鼠试验用莫沙必利的最佳剂量,本试验随机选用120只昆明系小鼠,分别用20 mg/kg·bw、10 mg/kg·bw、5 mg/kg·bw、2 mg/kg·bw的莫沙必利混悬液对小鼠进行粪便试验和肠推进试验,观察小鼠的首粒黑便时间,6 h和24 h排便量及重量,以及墨汁在小肠中的推进率。结果显示,小肠推进率试验中10 mg/kg·bw莫沙必利组与空白组比较有显著差异(P<0.05),各组与阳性对照组差异极显著(P<0.01);粪便试验中,10 mg/kg·bw莫沙必利组与药物空白组相比各项指标差异均为极显著(P<0.01),而且与模型空白组相比较除首粒黑便时间表现出极显著外(P<0.01),其余指标均表现为不显著,此结果与其余剂量组相比较有着较大优势。表明小鼠试验用莫沙必利剂量宜选用10 mg/kg·bw。 In order to screen the optimal dose for mosapride in mice,120 Kunming mice were randomly selected for the experiment.The mosapride suspension of 20 mg/kg·bw,10 mg/kg·bw,5 mg/kg·bw and 2 mg/kg·bw was subjected to a stool test and a bowel advancement test to observe the first black stool time,the 6 h and 24 h stool volume and weight,and the advancement rate of the ink in the small intestine.Results:The 10 mg/kg·bw mosapride group in the small intestine propulsion rate test was significantly different from the blank group(P<0.05).The difference between the groups and the positive control group was extremely significant(P<0.01).In the fecal test,The difference of the index between the 10 mg/kg·bw mosapride group and the drug blank control group was extremely significant(P<0.01),and compared with the model blank group,the time of the first black stool was extremely significant(P<0.01).The other indicators were not significant,and this result had a greater advantage than the other dose groups.The dose of mosapride in mice should be 10 mg/kg·bw.
