摘要
[目的]观察体积分数为10%的氢氧化钾溶液对成蚊永久性玻片标本制片效果的影响。[方法]分别采用5种标本处理方法:①未使用体积分数为10%的氢氧化钾;②使用体积分数为10%的氢氧化钾消化2h;③使用体积分数为10%的氢氧化钾消化4h;④使用体积分数为10%的氢氧化钾消化6h;⑤使用体积分数为10%的氢氧化钾消化8h。[结果]体积分数为10%的氢氧化钾消化时间6h或8h时,制作的标本镜下观察结构清晰;镜下(400×或100×)观察。显示,复眼由许多小眼构成,数量不定;正面观每个小眼呈现近似正六边形形状,与周围的六个小眼相邻,排列紧密,无缝隙,结构与蜂巢相似;侧面观小眼呈规则的泡状凸起,排列近似鱼鳞状。[结论]使用体积分数为10%的氢氧化钾溶液对成蚊消化6h或8h,制作的标本镜下观察结构清晰,适用于科研、教学。
[Objective]To observe the effect of 10% KOH solution on the preparation of permanent adult mosquito slide-sections. [Methods]5 groups with different kinds of sample-treatment methods were used: the control group(without 10% KOH solution), and the groups with 10% KOH solution treatment for 2 h, 4 h, 6 h and 8 h. [Results] The histological structures of adult mosquito under microscope was clear with 6 or 8 h treatments of 10% KOH solution. Results showed that the compound eyes were constructed by uncertain quantities of small eyes;At the front view, each small eye presents an approximately regular hexagonal shape and arranges closely and seamlessly adjacent to the surrounding six small eyes like honeycomb;At the lateral view, the small eyes showed as regularly bubble-like salients and arranged like fish scale. [Conclusion]The adult mosquito slide-sections with 10% KOH solution’s digestion of 6 h or 8 h has clear structures under microscope and is suitable for research and teaching.
