摘要
选用纤维素酶法制备海茸抗氧化多糖,并设计应用响应面法优化制备工艺。在单因素试验的基础上,选取pH、温度和料液比为自变量,以羟自由基清除活性及超氧阴离子清除活性为响应指标,确定纤维素酶法制备海茸抗氧化多糖的最佳工艺为料液比1∶40(g∶mL),加酶量2%,pH 5,在50℃水浴中搅拌提取2 h。在该试验条件下,超氧阴离子抑制率为(22.28±0.97)%,羟自由基抑制率为(77.64±1.82)%,试验值与预测值相近,说明模型拟合良好,优化工艺准确可行。
Cellulase-catalyzed hydrolysis was used to extract anti-oxidant polysaccharides from Durvillaea antarctica polysaccharides.Respone surface analysis methodology(RSM)was applied to optimize extraction process.Based on single-factor experiments,pH,extraction temperature and solid-liquid ratio were selected as affecting factors for Box-Behnken central composite experiment design,and hydroxyl radical scavenging activity and superoxide anion scavenging activity were taken as response indexes to determine the best process for preparation of anti-oxidant polysaccharides from Durvillaea antarctica by cellulase method.The process was as follows:the solid-liquid ratio was 1∶40(g∶mL),the cellulase dosage was 2%,the pH was 5,and the mixture was extracted in a water bath at 50℃for 2 h.Under the optimum conditions,the superoxide free radicals scavenging capacity was(22.28±0.97)%and the hydroxyl radical scavenging capacity was(77.64±1.82)%.The experimental values were close to the predicted values,which indicated that the model fit well and the optimization process was accurate and feasible.
