摘要
为探究与阿勒泰羊产羔数相关的DNA标记,本试验以153只产羔数不同的阿勒泰羊作为样本,利用PCR-SSCP、DNA测序等方法,对COIL、FSHR基因的遗传多样性进行研究,并分析其多态性与产羔数的相关性。结果显示,在阿勒泰羊中存在COIL基因(g.7321466G>C)、FSHR基因(g.75320741C>T)两个突变位点,均具有多态性。COIL基因产生3种基因型(GG、GC、CC),基因型频率分别为0.235、0.667、0.098,G为优势等位基因;FSHR基因产生3种基因型(CC、CT、TT),基因型频率分别为0.235、0.190、0.575。群体遗传学分析显示,COIL、FSHR基因在阿勒泰羊群体的PIC值分别为0.370、0.344,都属于中度多态位点。差异显著性分析结果表明,COIL基因GC、CC型的产羔数极显著高于GG型(P<0.01),GC型与CC型之间的产羔数差异不显著(P>0.05);FSHR基因3种基因型间的产羔数均差异不显著(P>0.05)。COIL基因g.7321466G>C位点有望作为提高阿勒泰羊产羔数的有效DNA标记。
The aim of this project was to explore DNA markers related to litter size of Altay sheep.In this experiment 153 Altay sheep with different litter size were taken as experimental samples,and methods such as PCR-SSCP and DNA sequencing were used to study the genetic diversity of COIL and FSHR genes,and to analyze the correlation between their polymorphism and litter size.The results showed that COIL gene(g.7321466G>C)and FSHR gene(g.75320741C>T)were found in Altay sheep with polymorphism.COIL gene produced three genotypes(GG,GC and CC),with genotype frequencies of 0.235,0.667 and 0.098,respectively,and G was the dominant allele.FSHR gene produced three genotypes(CC,CT and TT),with genotype frequencies of 0.235,0.190 and 0.575,respectively.Population genetic analysis showed that the PIC values of COIL and FSHR genes were 0.370 and 0.344,respectively,and both of them belonged to intermediate polymorphic sites.ignificant difference analysis results showed that the litter size of COIL gene with GC and CC type was significantly higher than that of GG type(P<0.01),while the litter size of GC type and CC type was not significantly different(P>0.05).There was no significant difference in lambing number among FSHR genotypes(P>0.05).In conclusion,the locus of COIL gene g.7321466G>C was expected to be an effective DNA marker to increase the litter size of Altay sheep.
