藏药三味宽筋藤汤散急性毒性试验及抗炎镇痛作用的研究

Study on the Acute Toxicity Test,Anti Inflammation and Analgesic Effect of Tibetan Medicine Sanwei Kuanjinteng Decoction Powder

目的:评价藏药三味宽筋藤汤散急性毒性以及抗炎镇痛的作用。方法:小鼠40只,雌雄各半,实验前禁食不禁水16 h,分别灌胃三味宽筋藤汤散最大浓度(1.15636 g原生药/mL),最大体积(0.4 mL/10 g),一天灌胃给药2次,间隔6 h,对照组灌胃等体积0.5%CMC,观察并记录动物在给药后14d内的中毒表现及死亡情况;以冰醋酸致小鼠腹腔通透性增加模型观察三味宽筋藤汤散的抗炎作用,以小鼠热板法和醋酸致小鼠扭体反应评价三味宽筋藤汤散的镇痛作用。结果:小鼠在灌胃给药后14 d内未出现明显的毒性表现,期间亦未出现动物死亡,解剖观察其主要脏器亦无明显异常发现;三味宽筋藤汤散3.0 g原生药/kg、1.0 g原生药/kg对醋酸致小鼠腹腔通透性升高具有一定的抑制作用,也有减少醋酸致小鼠扭体反应的趋势,但对热板法致小鼠疼痛反应无明显影响。结论:测得小鼠一次性口服三味宽筋藤汤散的最大耐受量为34.2 g原生药/kg,该剂量为临床拟用剂量(6.0 g/d,0.1 g/kg)的342倍,提示藏药三味宽筋藤汤散临床用药是比较安全的,同时具有一定的抗炎、镇痛作用。

Objective To evaluate the acute toxicity,anti-inflammatory and analgesic effects of the Tibetan medicine Sanwei kuanjinteng Decoction powder.Methods Forty mice,half male and half female,were fasted for 16 hours before the experiment.The maximum concentration(1.15636 g/mL)and the maximum volume(0.4 ml/10 g)of Sanwei kuanjinteng Decoction powder were respectively administered by gavage twice a day,with an interval of 6 hours.The volume of 0.5%CMC in the control group was administered by gavage.The poisoning performance and death of the animals within 14 days after the administration were observed and recorded.To observe the anti-inflammatory effect of Sanwei kuanjinteng Decoction powder by the model of increasing abdominal permeability induced by glacial acetic acid in mice and evaluation of analgesic effect of Sanwei kuanjinteng Decoction powder by hot plate method and writhing reaction induced by acetic acid in mice.Results There was no significant toxicity in the mice within 14 days after gavage,and no animal death occurred during the period.No significant abnormality was found in the main organs observed by anatomy.Sanwei kuanjinteng Decoction powder(3.0 g/kg,1.0 g/kg)has a certain inhibitory effect on the increase of abdominal permeability induced by acetic acid in mice,and also has a tendency to reduce the writhing response induced by acetic acid in mice,but has no significant effect on the pain response induced by hot plate method in mice.Conclusion The Tibetan medicine Sanwei kuanjinteng Decoction powder is safe in clinical use and has certain anti-inflammatory and analgesic effects.
