摘要
研究旨在提高菌株产中性蛋白酶的能力,采用单因素实验和正交实验,对一株高产中性蛋白酶的解淀粉芽孢杆菌进行了发酵培养基和发酵条件的优化,确定其最适培养基配方为玉米粉5%、棉粕3%、麸皮3%、KH2PO4 0.4%、Na2HPO4 0.05%、MgSO4 0.1%、MnCl2 0.01%,最适摇瓶培养条件为初始pH值7.0、种龄8 h、接种量5%、装液量100 ml/500 ml、30℃、240 r/min培养48 h,中性蛋白酶的摇瓶发酵酶活力最高可达16 826 U/ml,是初始发酵酶活力的3.98倍。
This experiment was conducted to improve the maximum enzyme activity of Bacillus amyloliquefaciens YF03. The enzyme-producing medium and conditions of Bacillus amyloliquefaciens YF03 were optimized by using single-factor experiment and orthogonal experiment. The optimum fermentation medium consisted of corn meal 5%, cottonseed meal 3%, wheat bran 3%,KH2PO4 0.4%, Na2HPO4 0.05%, MgSO4 0.1%, MnCl2 0.01%. And the best culture conditions was pH7.0,inoculum time 8 h, inoculum volume 5%, poured solution volume 100 ml/500 ml, shake speed 240 r/min, temperature 30 ℃, shake flask culture 48 h. Under the optimized conditions, the maximumenzyme activity reached 16 826 U/ml, which was 3.98 times compared with the activity before optimi-zation.
