肾衰泄浊汤及其拆方对周细胞-肌成纤维细胞转分化相关蛋白的影响 被引量:2

摘要 目的研究肾衰泄浊汤及其拆方对大鼠肾脏周细胞-肌成纤维细胞转分化(pericytes-myofibroblasts transition,PMT)相关蛋白表达的影响。方法取1个月龄健康雌性SD大鼠96只,随机平分为肾衰泄浊汤组、补虚方组、袪邪方组、贝那普利组、假手术组、模型组,每组16只,建立单侧输尿管梗阻(UUO)模型。随机取各组大鼠8只于术后7、14 d处死,取大鼠肾脏组织行HE、Masson染色,检测大鼠肾组织血小板衍生因子受体-β(PDGFR-β)、硫酸软骨素蛋白多糖(NG2)、α-平滑肌肌动蛋白(α-SMA)蛋白的表达。结果第7、14天时模型组、肾衰泄浊汤组、祛邪方组、补虚方组PDGFR-β、NG2、α-SMA表达高于假手术组(P <0.05);各治疗组PDGFR-β、NG2、α-SMA表达较模型组减少相对明显(P <0.05),肾衰泄浊汤组优于补虚方组、祛邪方组及贝那普利组。结论肾衰泄浊汤及其拆方可能是通过干预UUO大鼠PMT过程,抑制PDGFR-β、NG2、α-SMA表达,减轻肾间质纤维化,延缓慢性肾脏病的发展。 Objective To discuss the effect of Shenshuaixiezhuo decoction and its decoctions on theexpression of PMT related proteins in UUO rats.Methods One-month-old 96 healthy female SD rats were randomlydivided into Shenshuaixiezhuo decoction group,Buxufang group,Quxiefang group,benazepril group,sham opera-tion group,and model group,16 in each group. Establishing a UUO model. 8 rats in each group were randomlykilled at 7 and 14 days after operation. Renal tissues of rats were stained with HE and Masson to detect the expres-sion of PDGFR-β,NG2 and α-SMA protein.Results The expressions of PDGFR-β,NG2,and α-SMA in ratsin the model group,Shenshuaixiezhuo decoction group,Quxiefang group,and Buxufang group were higher thanthose in the sham operation group at day 7 and 14(P < 0.05);In comparison,the expressions of PDGFR-β,NG2,and α-SMA in the treatment groups were significantly reduced(P < 0.05). The Shenshuaixiezhuo decoction groupwas superior to the Buxufang group,Quxiefang group,and benazepril group.Conclusion Shenshuaixiezhuodecoction and its decoctions may inhibit the expression of PDGFR-β,NG2,α-SMA,alleviate renal interstitialfibrosis and delay the development of chronic kidney disease by intervening the PMT process of UUO rats.
