摘要
为确定土壤中乙蒜素的最佳提取方式,比较了甲醇、乙腈、二氯甲烷3种不同的提取溶剂和超声提取、加压流体萃取2种前处理方式提取土壤中乙蒜素的效果,用高效液相色谱法对提取液进行分析。结果表明,前处理方式采用超声提取效果优于加压流体萃取;极性较强的甲醇、乙腈提取乙蒜素平均回收率分别达到95.8%和96.2%,二氯甲烷提取平均回收率22.0%。对常温存放和密封冷藏2组样品连续5d测定乙蒜素含量,5d后常温存放的样品乙蒜素含量为初始值的14.9%,密封冷藏的样品乙蒜素含量为初始值的81.3%。
By comparing the effectiveness of three different solvents of methanol,acetonitrile and dichloromethane,as well as two pretreatment processes of ultrasonic and pressurised fluid extraction(PFE)to optimise the way for extracting ethylicin in soil,the extracts were analysed by high performance liquid chromatography(HPLC).It has shown that the effect of ultrasonic extraction was superior to that of PFE in the pretreatment,and the average recoveries of ethylicin extracted by strong polarity solvents methanol and acetonitrile could reach 95.8%and 96.2%,respectively,whilst that by dichloromethane would only be 22.0%.To determine the ethylicin contents in soil samples kept either at room temperature or by sealed cold storage after five days,that in the former one was only 14.9%left,whilst that in the latter one could be 81.3%remained.
