香砂六君子汤对功能性消化不良模型大鼠胃敏感性及十二指肠五羟色胺、TRPV1表达的影响 被引量:32

Effects of Xiangsha Liujunzi Decoction (香砂六君子汤) on Gastric Sensitivity and Expressions of 5-HT and TRPV1 in Duodenum of Functional Dyspepsia Model Rats

摘要 目的探讨香砂六君子汤治疗功能性消化不良的可能作用机制。方法 36只雄性SD大鼠随机分为空白组、模型组、香砂六君子汤组,每组12只。模型组和香砂六君子汤组采用碘乙酰胺灌胃+夹尾刺激法制备功能性消化不良大鼠模型。香砂六君子汤组予香砂六君子汤(浓度为0. 7875 g/ml) 1 ml/100 g灌胃,空白组、模型组予等量生理盐水灌胃,每日1次。7天后HE染色观察大鼠胃组织形态;检测大鼠胃敏感性;qPCR法检测大鼠十二指肠中瞬时感受器电位香草酸受体亚型1 (TRPV1) mRNA的含量;ELISA法检测大鼠十二指肠5-羟色胺(5-HT)含量;免疫荧光双标染色法检测大鼠十二指肠中TRPV1蛋白和5-HT的表达和分布。结果各组大鼠胃组织形态均正常。与空白组比较,模型组胃敏感性显著升高(P <0. 01),与模型组比较,香砂六君子汤组胃敏感性降低(P <0. 01);模型组大鼠十二指肠5-HT含量、TRPV1 mRNA表达较空白组显著升高(P <0. 01);与模型组比较,香砂六君子汤组大鼠十二指肠5-HT含量、TRPV1 mRNA表达明显下降(P <0. 01);TRPV1蛋白与5-HT共表达于十二指肠,模型组TRPV1蛋白和5-HT表达较空白组增多,而香砂六君子汤组表达较模型组减少。结论香砂六君子汤能下调功能性消化不良模型大鼠十二指肠TRPV1和5-HT的表达水平,从而缓解胃高敏状态,可能是其治疗功能性消化不良的作用机制之一。 Objective To explore possible mechanism of Xiangsha Liujunzi Decoction(香砂六君子汤,XSLJZ) in treating functional dyspepsia(FD). Methods A total of 36 male Sprague-Dawley(SD) rats were randomly divided into blank group,model group and XSLJZ group,with 12 rats in each group. The model group and the XSLJZ group were administrated with iodoacetamide by gavage combined with tail clamping to establish FD rat models. The XSLJZ group was given 1 ml/100 g(concentration of 0. 7875 g/ml) XSLJZ by gavage,while the blank group and the model group were given equal volume of normal saline by gavage,once a day. After 7 days,gastric tissue morphology was observed by HE staining,and rat’s gastric sensitivity was detected. Transient receptor potential vanilloid type 1(TRPV1) mRNA content in duodenum was detected by q PCR,and 5-hydroxytryptamine(5-HT) content in duodenum was detected by ELISA. Immunofluorescent double labelled staining was applied to detect expressions and distributions of TRPV1 protein and 5-HT in duodenum. Results The gastric histomorphology in the 3 groups was normal. Compared with the blank group,the gastric sensitivity was significantly increased in the model group(P <0. 01). Compared with the model group,the gastric sensitivity was decreased(P < 0. 01). The expression of TRPV1 mRNA and the content of 5-HT in duodenum in the model group were significantly increased compared with the blank group(P < 0. 01). Compared with the model group,the expression of TRPV1 mRNA and the content of 5-HT in duodenum in the XSLJZ group were significantly decreased(P < 0. 01). The TRPVl protein were expressed in the duodenum along with 5-HT,and the expressions of TRPV1 protein and 5-HT in the model group were more than those in the blank group,while the expression in the XSLJZ group were decreased compared with those in the model group.Conclusion XSLJZ can reduce expression levels of duodenal TRPV1 and 5-HT in FD rats to relieve gastric hypersensitivity,which may be one of its mechanisms in treating FD.
