摘要
目的研究Wnt通路中卷曲蛋白4(Fz4)和分泌型卷曲相关蛋白1(sFRP1)在急性淋巴细胞白血病(ALL)患儿的表达及作用。方法随机选取50例初发且经诱导治疗第33天骨髓达完全缓解的ALL患儿,分别留取化学药物治疗(简称化疗)治疗前骨髓和治疗第33天完全缓解骨髓,作为初发组和缓解组,收集20例免疫性血小板减少症患儿骨髓作为对照组,依照危险度分层将初发组50例ALL患儿划分高危组(n=12)、中危组(n=11)和低危组(n=27)。Real-time PCR法检测骨髓单个核细胞中Fz4 mRNA和sFRP1 mRNA表达,Western blot法检测骨髓单个核细胞中Fz4和sFRP1蛋白表达。结果初发组Fz4 mRNA和蛋白相对表达量明显高于缓解组和对照组(P<0.05)、sFRP1 mRNA和蛋白相对表达量明显低于缓解组和对照组(P<0.05)。初发组中Fz4与sFRP1的mRNA表达、蛋白表达均呈负相关(P<0.05)。高、中危组Fz4 mRNA和蛋白相对表达量均高于低危组(P<0.05),高、中危组sFRP1表达均低于低危组(P<0.05)。结论Fz4表达升高和(或)sFRP1表达降低可能参与儿童ALL的发病,其表达异常程度可能与预后相关。
Objective To investigate the expression of frizzled 4(Fz4)and secreted frizzled-related protein 1(sFRP1)in children with acute lymphoblastic leukemia in Wnt pathway and the impact on prognosis.Methods Totally 50 children with ALL newly diagnosed and achieving complete remission on the 33rd day after induction therapy was selected randomly.The bone marrow of these children with pre-treatment and CR were taken as the initial group and the remission group respectively.The bone marrow of 20 children with ITP was collected as the control group.According to risk stratification,50 children with ALL in the initial group were divided into high risk group(n=12)and intermediate risk group(n=11)and low risk group(n=27).Real-time PCR was used to measure the expression of Fz4 and sFRP1 mRNA,Western blot was used to detect the expression of Fz4 and sFRP1 protein.Results The expression of Fz4 mRNA and protein in the initial group was significantly higher than that in the remission group and the control group(P<0.05).The expression of sFRP1 mRNA and protein in the initial group was significantly lower than that in the remission group and the control group(P<0.05).There was a negative correlation between the expression of Fz4 and sFRP1 in the initial group(P<0.05).The expression of Fz4 mRNA and protein in the HR group and IR group was higher than that in the LR group(P<0.05).The expression of sFRP1 mRNA and protein in the HR group and IR group was lower than that in the LR group(P<0.05).Conclusion Increased expression of Fz4 and(or)decreased expression of sFRP1 may be involved in the pathogenesis of ALL of childhood,and the degree of abnormality of two factor′sexpression may be related to prognosis.
