龙眼叶抗氧化和抑制α-葡萄糖苷酶活性的谱效关系研究 被引量:9

Spectrum-effect Relationship between the Antioxidant and Inhibitory Activity ofα-Glucosidase in the Leaves of Dimocarpus longan

摘要 目的:研究龙眼叶不同极性部位HPLC色谱图与其体外抗氧化、抑制α-葡萄糖苷酶活性作用的谱效关系。方法:采用灰色关联度分析和Pearson相关分析法,研究龙眼叶不同极性部位清除DPPH自由基、抑制α-葡萄糖苷酶活性作用与HPLC色谱峰之间的相关性。结果:龙眼叶各极性部位对DPPH自由基均有明显清除作用,并能显著抑制α-葡萄糖苷酶活性。灰关联分析结果表明对抗氧化作用贡献较大的是1、18、22(槲皮素)号峰,对酶活性抑制贡献较大的是5、6、7、8、16号峰;而Pearson相关分析发现与抗氧化相关性较强的是14(没食子酸乙酯)号峰,与酶活性抑制相关性较强的是5、7、8、16、18号峰。结论:龙眼叶具有的抗氧化和抑制α-葡萄糖苷酶活性作用是多种成分共同作用的结果,可为中药谱效分析研究提供参考。 Objective:To study the spectrum-effect relationship between the HPLC chromatogram of different polar parts of Dimocarpus longan leaves and its antioxidant and inhibitory activities ofα-glucosidase in vitro.Methods:The correlation between scavenging activity on DPPH free radicals andα-glucosidase inhibiting activity of different polar parts and the chromatographic peaks of HPLC was studied by gray correlation degree analysis and Pearson correlation analysis.Results:Different polar parts of Dimocarpus longan leaves had an obvious scavenging effect on DPPH free radical and could obviously inhibit the activity ofα-glucosidase.The results of grey correlation analysis showed that peaks 1,18 and 22(quercetin)had relatively large contribution on the antioxidant function,the peaks 5,6,7,8 and 16 had great contribution on enzyme activity inhibition.While peak 14(ethyl gallate)had relatively strong correlated with oxidant resistance through Pearson correlation analysis and peaks 5,7,8,16 and 18 had relatively strong correlated with enzymatic activity inhabitation.Conclusion:The antioxidant and inhibitory action ofα-glucosidase activity of Dimocarpus longan leaves are the results of the combined action of various components,which can provide reference for the spectrum-effect analysis of traditional Chinese medicine.
