摘要
目的:优选10种大孔树脂富集纯化延龄草中3种甾体皂苷的工艺研究。方法:采用HPLC法,以重楼皂苷Ⅵ、偏诺皂苷元-3-O-α-L-吡喃鼠李糖基-(1→4)-[α-L-吡喃鼠李糖基-(1→2)]-β-D-吡喃葡萄糖苷、重楼皂苷Ⅶ的含量为指标,考察上样浓度、洗脱溶剂及用量、不同树脂类型对3种甾体皂苷吸附和转移率的影响。结果:优选AB-8型大孔树脂,取14 g延龄草粉末制备上样供试液,流速2 BV/h,蒸馏水洗脱8 BV,30%乙醇洗脱7 BV,60%乙醇2 BV洗脱除杂,85%乙醇6 BV富集甾体皂苷成分。结论:优选工艺可较好地富集延龄草3种甾体皂苷,简单易行,生产成本低。
Objective:To optimize the process of enriching and purifying three steroidal saponins from Trillium tschonoskii by ten kinds of macroporous resins.Methods:HPLC method was used to determine the contents of polyphyllinⅥ,pennogenin-3-O-α-L-rhamnopyranoside(1→4)-[α-L-rhamnopyranoside(1→2)]-β-D-glucopyranoside,polyphyllinⅦ.The effects of sample concentration,elution solvent and dosage and different resin types on the adsorption and transfer rate of three steroidal saponins were investigated.Results:AB-8 macroporous resin was selected.14 g powder of Trillium tschonoskii was used to prepare the test solution,and the flow rate was 2 BV/h,distilled water eluated 8 BV,30%ethanol eluated 7 BV,60%ethanol eluated 2 BV.Steroidal saponins were enriched with 85%ethanol for 6 BV.Conclusion:The optimized separation process can enrich three steroidal saponins from Trillium tschonoskii.It is simple and feasible,the production cost is low.
