摘要
目的:研究白藜芦醇对血小板源性生长因子(PDGF)-BB诱导的血管平滑肌细胞(VSMCs)葡萄糖转运蛋白4(GLUT4)转位的抑制作用及机制。方法:不同浓度白藜芦醇预处理后用PDGF-BB刺激VSMCs;提取膜蛋白,Western blot检测膜上GLUT4表达;细胞免疫荧光法检测GLUT4膜转位;葡萄糖摄取荧光探针(2-NBDG)检测细胞葡萄糖摄取;Western blot检测白藜芦醇预孵育对Akt磷酸化的影响。结果:10 ng/mL PDGF-BB能诱导VSMCs GLUT4膜转位和葡萄糖摄取,白藜芦醇能浓度依赖性抑制PDGF-BB引起的GLUT4膜转位和葡萄糖摄取,并能抑制PDGF-BB诱导的Akt磷酸化。结论:白藜芦醇能抑制PDGF-BB诱导的VSMCs中GLUT4膜转位和葡萄糖摄取,其机制可能与对Akt磷酸化的抑制有关。
Objective:To investigate the inhibitory effect and mechanism of resveratrol on platelet-derived growth factor(PDGF)-BB-induced translocation of glucose transporter 4(GLUT4)in vascular smooth muscle cells(VSMCs).Methods:VSMCs were stimulated by PDGF-BB after pretreatment with different concentrations of resveratrol.Membrane proteins were extracted and GLUT4 expression was detected by Western blot;GLUT4 membrane translocation was detected by cellular immunofluorescence assay;Glucose uptake was detected by 2-NBDG;The effect of resveratrol pre-incubation on Akt phosphorylation was detected by Western blot.Results:10 ng/mL PDGF-BB could induce GLUT4 translocation and glucose uptake in VSMCs.Resveratrol could inhibit GLUT4 translocation and glucose uptake caused by PDGF-BB in a concentration-dependent manner,inhibit Akt phosphorylation induced by PDGF-BB.Conclusion:Resveratrol can inhibit GLUT4 membrane translocation and glucose uptake in VSMCs induced by PDGF-BB,the mechanism may be related to the inhibition of Akt phosphorylation.
