不同剂型剂量黄连对糖尿病大鼠血糖及肠道二糖酶活性的影响研究 被引量:1

摘要 目的:观察不同剂量剂型的黄连制剂对2型糖尿病(T2DM)大鼠血糖、胰岛素抵抗及肠道二糖酶活性的影响。方法:采用高脂高糖饲料联合40mg﹒kg^-1 链脲佐菌素腹腔注射的方法建立 T2DM 大鼠模型。90只雄性SD大鼠随机分为以下9组,每组10只,分别是:空白组、模型组、阿卡波糖组、汤剂、浸渍剂和粉剂高低剂量组,给药2周后,观察大鼠一般情况、空腹血糖、胰岛素及肠道二糖酶活性。结果:除黄连汤剂低剂量组外,其余各治疗组就能降低大鼠空腹血糖值、减轻胰岛素抵抗(均P<0.05)。黄连可抑制糖尿病大鼠模型中的肠道二糖酶活性,以浸渍剂高剂量组对肠道二糖酶的作用最广泛。结论:不同剂型剂量黄连对血糖和肠道二糖酶活性的影响不同,总体趋势是高剂量组优于低剂量组;降糖效果以粉剂性价比最高,对肠道二糖酶的抑制作用以浸渍剂效果最好。 Objective:This article is to investigate the effect of Rhizoma Coptidis (RC) with different doses and dosage forms on blood glucose,insulin resistance and activities of intestinal disacchridases in type 2 diabetic rats.Method:The rat model was established by feeding with high fat and sugar diet and then being injected with peritoneal streptozotocin (STZ,40 mg﹒kg^-1).90 male SD rats were randomly divided into the following nine groups with 10 in each group:blank group,model group,acarbose group,low-dose and high-dose group of RC decoction,RC soaking group and RC powder group respectively.After two weeks of oral gavages,the general condition of rats were observed.The fasting blood glucose,insulin and intestinal disaccharidase activity were measured.Result:Compared with the model group,rats in the treatment group except those in the low-dose RC decoction group presented with markedly lower blood glucose level and lower insulin resistance index (P<0.05).Treatment with different doses and dosage forms of RC and acarbose reversed the increase in disacchridases activity.Among which,high doses of RC soaking form exhibited a general inhibitory effect on intestinal disaccharidase activity.Conclusion:High dose is better than low dose in terms of blood-lowering effect for RC.RC powder is recommended to get satisfactory blood-lowering result in respect to cost-effectiveness while soaking form is recommended in terms of better inhibitory effect on intestinal disacchridases activity.
