摘要
目的:探究补阳还五汤对急性脊髓损伤(SCI)大鼠NOD样受体蛋白1(NLRP1)/半胱氨酸蛋白水解酶-1(Caspase-1)细胞焦亡通路的影响。方法:将32只SPF级大鼠随机分成假手术组、甲强龙组、补阳还五汤(BYHWT)组和模型组,造模成功后补阳还五汤组每天给予补阳还五汤灌胃1次,连续14d,甲强龙组给予甲强龙冲击治疗;假手术组及模型组注射等补阳还五汤剂量的生理盐水;干预后的第1,3,7及14天以BBB评分评价大鼠神经功能恢复情况,Western Blot法检测损伤脊髓中组织NLRP1,Caspase-1及IL-18蛋白的表达。结果:BBB评分:假手术组大鼠评分无变化,BYHWT组及甲强龙组治疗后3d,7d及14d评分大于模型组,差异有统计学意义(P<0.05),治疗后7d及14d,各组大鼠评分变化不明显。干预后第14天大鼠损伤脊髓组织中NLRP1,Caspase-1及IL-18蛋白的表达结果:BYHWT组和甲强龙组蛋白表达均低于假手术组,高于模型组,差异有统计学意义(P<0.05)。结论:补阳还五汤联合脊髓减压治疗急性SCI效果明显,作用机制可能是通过抑制NLRP1/Caspase-1通路以减少细胞焦亡。
Objective:To investigate the effect of Buyanghuanwu decoction(BYHWT)on pyroptosis pathway of NLRP1(NOD-like receptor protein 1)/Caspase-1(cysteinyl aspartate specific proteinase)in rats with acute spinal cord injury(SCI).Methods:Thirty-three SPF rats were randomly divided into four groups:sham operation group,methylprednisolone group,BYHWT group and model group.After the model was established successfully,BYHWT was prepared into a decoction once a day for 14 d.The methylprednisolone group was treated by methylprednisolone pulse therapy.The rats in the sham operation group and the model group were given normal saline at the dose of Buyanghuanwu decoction.The neurological recovery of the rats was evaluated by BBB scale at the 1,3,7,14 dafter intervention.The protein expression of NLRP1,Caspase-1 and IL-18 in SCI tissue was detected by Western Blot method.Results:BBB scale:There was no change in the scale of rats in the sham operation group.The scale of BYHWT group and methylprednisolone group were higher than that of the model group at the 3,7 and 14 dafter treatment,the differences were statistically significant(P<0.05).At the 7 and 14 dafter treatment,there was no significant change in the scale of rats in each group.The protein expression of NLRP1,Caspase-1 and IL-18 in SCI tissue of rats at the 14 dafter intervention:the protein expression of NLRP1,Caspase-1 and IL-18 in BYHWT group and methylprednisolone group were significantly lower than those in sham operation group and significantly higher than those in model group,the differences were statistically significant(P<0.05).Conclusion:The therapeutic effect of Buyanghuanwu decoction combined with spinal cord decompression on acute SCI is obvious,and the mechanism may be to reduce pyroptosis death by inhibiting NLRP1/Caspase-1 pathway.
