摘要
目的:对地龙提取物抗肺纤维化的有效成分进行分析。方法:将60只小鼠随机分为正常对照组、模型组和地龙组。用暴露式气管内注入法造模后,于7 d、28 d处死动物,测定其肺指数、肺病理形态变化及肺组织羟脯氨酸(HYP)含量。结果:地龙组与模型组比较,肺指数明显降低(P<0.05);病理结果明显好于模型对照组;光镜下,正常对照组肺组织结构清析,无明显炎症;其他两组在7 d时均出现明显的炎症反应,大量炎细胞浸润,细胞间隔水肿增厚,各组间镜下差异不明显;28 d除正常对照组外,各组均有不同程度的纤维化改变,纤维组织增生间隔增宽,模型对照组尤为严重,而地龙组显示轻度纤维化改变。HYP含量方面比较,7 d时,模型组与对照组相比差异明显(P<0.05),28 d时,地龙组和正常对照组比较差异明显,具有统计意义(P<0.05),与模型组比较也具有统计学意义(P<0.05)。结论:地龙提取物可不同程度改善博莱霉素所致的小鼠肺纤维,能够为临床上针对不同类型的肺纤维化患者的合理使用提供数据支持。
Objective:To analyze the effective components of earthworm extract against pulmonary fibrosis.Methods:60 mice were randomly divided into the normal control group,the model group and the earthworm extract group.The exposure endotracheal injection method was used to make the model of pulmonary fibrosis.The animals were sacrificed on day 7th and day 28th,and the lung index,pulmonary morphological changes and HYP content in lung tissues were measured.Results:Compared to the model group,lung index was significantly reduced in the earthworm extract group(P<0.05).The pathological results were better in the earthworm extract group than those in the model group.Under the light microscope,the lung tissue structure of the normal control group had no obvious inflammation.The other groups all showed obvious inflammatory response on day 7th,with a large number of inflammatory cells infiltrating and cell septal edema thickening,and there were no significant difference between each group under the microscope.After 28 days,except the normal control group,the other groups had different degree of fibrosis change,and the fibrous tissue hyperplasia interval was widened,the model control group was especially severe,the other groups showed mild fibrosis change.In terms of HYP content,on day 7th,the difference between the model group and the control group was significant(P<0.05);on day 28th,there were statistical differences among the model group,the earthworm extract group and the normal control group(P<0.05).Conclusion:The extract of earthworm can improve the pulmonary fibers induced by bleomycin in mice to varying degrees,which can provide data support for its rational application to different types of pulmonary fibrosis.
