云南野生大红菇(Russula vinosa)红色素的制取、抗氧化性研究及其成分分析 被引量:4

摘要 该文研究并优化了云南产野生大红菇(Russula vinosa)子实体红色素的提取条件,评价了红菇红色素提取物的稳定性和抗氧化活性,采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱(ultraperformance liquid chromatography-quadrupole-orbitirp-mass spectrometry,UPLC-Q-Orbitrap-MS)对提取物中的主要物质成分进行了分析和鉴定。结果表明,红菇子实体红色素的最佳提取工艺条件为乙醇体积分数40%,料液比1∶90(g∶m L),"超声5 min,静置5 min,涡旋振荡30 s"循环5次;红菇色素提取物在一定的加热条件、p H环境、紫外光线照射、金属离子作用以及食品添加剂作用下可保持较好稳定性。红菇色素提取物对DPPH·、ABTS+·、·OH具有一定的清除作用,对铁离子(Fe3+)具有一定的还原作用,并较好地抑制脂质体过氧化的能力。通过质谱分析,鉴定了红菇色素提取物中的31个化学成分,其中有机酸类9个、多元醇类1个、多糖类1个、维生素类1个、花青素类9个,确定了红菇红色素为水溶性花色苷类色素。 The extraction conditions of red pigments from Russula vinosa in Yunnan province were optimized by orthogonal test. The stability and antioxidant activities of the pigment extracts were evaluated,and the main components were analyzed and identified by UPLC-Q-Orbitrap-MS system. The results showed that the optimum extraction conditions were ethanol concentration of 40%,solid to liquid ratio of 1∶ 90( g∶ m L),and the 5 cycles of"ultrasonic treatment for 5 min,stand for 5 min,vortex oscillation for 30s". The pigment extracts exhibited good stability under certain heating conditions,and different pH ranges,with ultraviolet light irradiation,metal ions and food additives. The pigment extracts showed scavenging abilities on DPPH·,ABTS+· and ·OH,reducing effect on Fe3+,and inhibiting activity on lipid peroxidation. According to the MS analysis,31 components were identified from the pigment extracts,including 9 organic acids,1 polyol,1 polysaccharide,1 vitamin and 9 anthocyanins. The red pigment of R. vinosa was identified as water-soluble anthocyanins.
