摘要
目的:研究黄腐酚对胆管癌QBC939细胞生长和凋亡的影响及作用机制。方法:将人胆管癌QBC939细胞培养在细胞培养板中,加入不同浓度的黄腐酚(0μM、10μM、20μM、30μM),药物作用后24 h收集细胞。应用CCK-8法检测细胞存活率。AnnexinⅤ-FITC/PI双染后用流式细胞仪检测细胞凋亡率。免疫荧光法检测Cleaved Caspase-3蛋白的表达。Western blot检测Notch1、NF-κB p65、cleaved Caspase-3蛋白的表达。结果:黄腐酚(10μM、20μM、30μM)能够明显抑制胆管癌QBC939细胞生长。黄腐酚可上调Caspase-3活性裂解片段的表达,诱导胆管癌细胞凋亡。此外,黄腐酚可抑制Notch1、NF-κB p65的表达。结论:黄腐酚对胆管癌QBC939细胞有较强的生长抑制和凋亡诱导作用,其机制可能与抑制Notch1-NF-κB通路有关。
Objective:To investigate the mechanism of antiproliferative and apoptotic effect on human Cholangiocarcinoma QBC939 cells by Xanthohumol.Methods:The QBC939 cells were cultured in cell cultrue plate and then treated with 10μM,20μM,30μM xanthohumol or DMSO(control).CCK-8 assay was used to assess the viability of cells after xanthohumol treatment.The apoptosis induced by xanthohumol was detected by Flow cytometry using AnnexinⅤ-FITC/PI Apoptosis Detection Kit.The effect of xanthohumol on the cleaved caspase-3 expression was determined by immunoflfluorescence confocal microscopy.The effect of xanthohumol on the expression of Notch1、NF-κB p65、cleaved caspase-3 was determined by Western blot.Results:Xanthohumol(10μM,20μM,30μM)inhibited the proliferation of human cholangiocarcinoma QBC939 cells in a dose-dependent manner.Xanthohumol increased the expression of cleaved caspase-3 and induced cell apoptosis.Furthermore,the expressions of Notch1、NF-κB p65 were decreased by xanthohumol.Conclusions:Notch1-NF-κB cell signaling pathway could be involved in xanthohumol-induced proliferation inhibition and apoptosis in human cholangiocarcinoma QBC939 cells.
