摘要
目的:基于网络药理学研究茶枯提取物的抗血栓作用机制。方法:选择茶枯提取物中16个活性成分,通过Pharm Mapper网络平台预测潜在靶点,通过Gene cards及OMIM网络平台检索血栓相关基因,将两者交集获取茶枯提取物活性成分抗血栓靶点,进而构建PPI网络、活性成分-交集基因-疾病网络,对GO及KEGG通路进行富集分析。结果:共交集得到91个茶枯提取物活性成分抗血栓靶点,主要涉及AKR1B1、MME、CASP3、F2和HSP90AA1。83个GO条目全部涉及分子功能。99条KEGG通路中P值最显著的前5个依次为:流体剪切应力与动脉粥样硬化、催乳素信号途径、百日咳、胰岛素抵抗及补体及凝血级联反应。结论:茶枯提取物抗血栓作用机制涉及多种成分及多个靶点。
Objective:To study the antithrombotic mechanisms of Camellia cake extracts by network pharmacology.Methods:16 active ingredients were selected from in Camellia cake extracts,potential targets were predicted by Pharm Mapper,search thrombus-related genes were searched from through Gene cards and OMIM,then intersection targets were obtained,PPI network,active ingredient-intersection gene-disease network were constructed,enrichment analysis of GO and KEGG pathway was performed.Results:91 antithrombotic targets of active ingredients were obtained,which were mainly involved with AKR1B1,MME,CASP3,F2,and HSP90AA1.All 83 GO entries were related to molecular functions.The top 5 KEGG pathways with most significant p-values were:fluid shear stress and atherosclerosis,prolactin signaling pathway,pertussis,insulin resistance,and complement and coagulation cascades.Conclusions:The antithrombotic mechanisms of Camellia cake extracts might involve multiple components and multiple targets.
