摘要
目的:通过层层自组装修饰磁性二硫化钨(magnetic WS 2,mWS 2)构建生理条件下稳定的药物负载平台,靶向输送药物并考察光热化疗协同对乳腺癌MCF-7细胞株的杀伤作用。方法:利用溶剂热法制备mWS 2,并在水体系中逐层包裹带正电的壳聚糖(chitosan,CS)和带负电的羧甲基纤维素钠(carboxymethyl cellulose sodium,CMC),制备mWS 2-CS/CMC复合材料。对所合成材料进行系列结构表征,并研究材料的光热性能以及药物的负载、释放行为。设计体外细胞光热化疗联合治疗方案,同时考察其细胞摄取和细胞毒性。结果:合成的mWS 2-CS/CMC具有超顺磁性且稳定性好,载药量高,药物释放表现出对pH和近红外激光的双响应释放曲线。此外,毒性低且可以被充分摄取,对乳腺癌MCF-7细胞株的光热化疗表现出优异的治疗效果。结论:制备的mWS 2-CS/CMC为一种优异的药物载体,在光热治疗以及光热化疗协同治疗领域有望得到应用。
Objective:Layer-by-layer self-assembly was used to modify magnetic tungsten disulfide(mWS 2),thus a stable drug loading platform under physiological conditions was built.It had a targeted drug delivery property and synergistic effect on cancer cells through photo-chemotherapy.Methods:mWS 2 was prepared by solvothermal method,and then positively charged chitosan and negatively charged sodium carboxymethyl cellulose were layer-by-layer deposited in the water system respectively to prepare chitosan/carboxymethyl cellulose sodium modified magnetic tungsten disulfide composite material(mWS 2-CS/CMC).Series of structural characterizations was performed on the synthesized materials and,and its photothermal properties as well as its drug loading and release behavior was studied.In addition,in vitro cell photochemotherapy combined treatment plan was designed,at the same time to its cell uptake and cytotoxicity was examined.Results:The synthesized mWS 2-CS/CMC was superparamagnetic with good stability and high drug loading profile.The drug release assay showed a dual response release to pH and NIR.In addition,it has low toxicity and can be taken up by MCF-7 cells,and produce an excellent therapeutic effect on the combination of photo-chemotherapy on cancer cells.Conclusion:The mWS 2-CS/CMC is an excellent drug carrier and can also be used as a photothermal agent for photothermal therapy and combination therapy.
