摘要
目的建立能同时测定丹桂香颗粒中芍药苷、丹皮酚和丹参酮ⅡA 3种活性成分含量的RP-HPLC法。方法丹桂香颗粒研细后,加甲醇超声提取;乙腈-3 mL·L^-1磷酸溶液为流动相,梯度洗脱;Waters Xbridge RP-C 18色谱柱(250 mm×4.6 mm,5μm);检测波长:247 nm;柱温:30℃;进样量:25μL;以标准曲线法定量检测。结果芍药苷、丹皮酚和丹参酮ⅡA的平均回收率分别为97.9%,98.2%和94.0%;均在质量浓度1.0~50.0μg·mL^-1范围内线性关系良好;r值均大于0.9950;精密度和重复性RSD值均小于5.0%(n=6)。结论该方法具有简便、快速、结果准确等优点,适用于丹桂香颗粒的质量控制。
Objective To establish an RP-HPLC method for determining the contents of 3 active ingredients paeoniflorin,paeonol and tanshinoneⅡA in Danguixiang Granules.Methods Danguixiang Granules samples were grinded first and then extracted with methanol.Acetonitrile-3 mL·L^-1 phosphoric acid solution as gradient elution solvents,samples were separated on Waters Xbridge RP-C 18 column(250 mm×4.6 mm,5μm)at 30℃and detected at 247 nm with injection volume of 25μL.Standard curve method was used for the quantitative analysis.Results Paeoniflorin,paeonol and tanshinoneⅡA showed good linear relationships in the range of 1.0-50.0μg·mL^-1 with correlation coefficients(r)all above 0.9950.Average recoveries of paeoniflorin,paeonol and tanshinoneⅡA were 97.9%,98.2%and 94.0%,respectively.RSDs of precision and repeatability were all below 5.0%(n=6).Conclusion The method is simple,quick and accurate,which is suitable for the quality control of Danguixiang Granules.
