摘要 目的:检测7-二氟亚甲基-5,4’-二甲氧基染料木黄酮(DFOG)对H2O2氧化应激人子宫螺旋动脉内皮细胞(HUSAEC)增殖凋亡的影响。方法:体外培养HUSAEC细胞,在加入H2O2氧化应激诱导前30 min,分别添加不同浓度DFOG(2、4、8μmol/L)进行预孵,MTT检测DFOG对H2O2氧化应激诱导的HUSAEC细胞的增殖促进作用,FCM检测DFOG对H2O2氧化应激诱导的HUSAEC细胞凋亡情况。结果:与H2O2组比较,DFOG各组明显减轻了H2O2对HUSAEC细胞增殖的抑制作用,减少H2O2氧化应激诱导HUSAEC细胞的凋亡(P<0.05),呈浓度和时间依赖性。结论:DFOG对H2O2氧化应激HUSAEC细胞有明显的促进增殖及抑制凋亡作用,提示DFOG可能成为妊娠期高血压疾病治疗的新型药物。 Objective:To determine the effect of 7-difluoromethy-5,4’-dimethoxy genistein(DFOG)on proliferation and apoptosis of human uterine spiral artery endothelial cells(HUSAEC)induced by H2O2 oxidative stress.Method:HUSAEC cells were cultured in vitro,30 minutes before the induction of H2O2 oxidative stress,different concentrations of DFOG(2,4,8μmol/L)were added for pre incubation.MTT was used to detect the effect of DFOG on the proliferation of HUSAEC cells induced by H2O2 oxidative stress,FCM was used to detect the effect of DFOG on the apoptosis of HUSAEC cells induced by H2O2 oxidative stress.Result:Compared with H2O2 group,DFOG group significantly reduced the inhibition of H2O2 on the proliferation of HUSAEC cells,and reduced the apoptosis of HUSAEC cells induced by H2O2 oxidative stress(P<0.05),which was concentration and time-dependent.Conclusion:DFOG can significantly promote proliferation and inhibit apoptosis of H2O2 oxidative stress HUSAEC cells,suggesting that DFOG may be a new drug for the treatment of hypertension during pregnancy.
