摘要 目的:探究miR-146a对非小细胞肺癌细胞系A549的增殖及迁移的影响,进一步研究miR-146a的靶向基因及其作用。方法:miR-146a mimics转染非小细胞肺癌A549细胞;CCK8及Transwell检测过表达miR-146a对非小细胞肺癌细胞系A549增殖及迁移的影响;利用Targetscan预测及荧光素酶报告基因系统确认miR-146a可能结合的靶基因;反转录聚合酶链式反应(reverse transcription-polymerase chain reaction,RT-PCR)及Western Blot检测过表达miR-146a对靶基因的影响。结果:过表达miR-146a显著抑制非小细胞肺癌细胞系A549增殖(P<0.001)及迁移(P<0.001);miR-146a结合的潜在靶基因为Smad4;过表达miR-146a显著上调其靶基因Smad4基因的表达(P<0.05)。结论:miR-146a通过上调其靶基因Smad4抑制非小细胞肺癌细胞系A549的增殖及迁移。 Objective:To explore the effect of miR-146a on the proliferation and migration of non-small cell lung cancer cell line A549,and further to study the target gene of miR-146a and its role.Methods:Reverse transcription-polymerase chain reaction(RT-PCR)was used to detect expression of miR-146a in non-small cell lung cancer A549 cells transfected with miR-146a mimics;CCK8 and transwell was used to detect effect of overexpression of miR-146a on proliferation and migration of non-small cell lung cancer cell line A549;target scan and luciferase reporter gene system was used to detect confirm the binding target genes for miR-146a;RT-PCR and Western Blot were used to detect the effect of overexpression of miR-146a on target genes.Results:Overexpression miR-146a significantly inhibits proliferation(P<0.001)and migration(P<0.001)of non-small cell lung cancer cell line A549;the potential target gene for miR-146a is Smad4;overexpression miR-146a significantly increase the expression of its target gene Smad4(P<0.05).Conclusion:MiR-146a inhibits proliferation and migration of non-small cell lung cancer cell line A549 by targeted up-regulation of Smad4.
