摘要
目的研究CD147基因过表达对皮肤毛发和创伤修复的影响。方法测量观察CD147基因过表达小鼠及同窝野生型对照小鼠生长发育状况;取1、5、8个月龄小鼠,随机拔取背部毛发,显微镜下分类、测量长度;选择8周龄野生型小鼠(WT)和CD147过表达转基因鼠各6只,进行9轮脱毛后,皮肤镜扫描检测毛发密度,背部打孔观察皮肤创伤愈合能力,冷冻切片后免疫荧光染色检测皮肤干细胞标志基因CD34表达情况和BrdU法观察皮肤细胞增殖情况。结果CD147过表达对小鼠生长发育和皮肤结构无显著影响;与同窝野生型小鼠毛发相比,各类型毛发长度差异无统计学意义,但分类统计显示各型毛发比例有变化,刚直毛比例减少,Z型绒毛增多;多轮脱毛后,CD147过表达小鼠毛发密度降低,创伤愈合速度减慢;免疫荧光显示CD147过表达小鼠皮肤毛囊干细胞增殖增加。结论CD147在皮肤毛发形成中发挥重要作用,其表达促进皮肤干细胞增殖,多轮脱毛后加速毛囊干细胞消耗,从而降低皮肤毛发密度和创伤愈合能力。
【Objective】To observe the effect of CD147 gene overexpression on skin hair regeneration and wound healing.【Methods】The basic phenotype such as growth of CD147 overexpressed mice and wild-type(WT)control mice were measured and observed.The dorsal hair of 1,5 and 8-month-old mice were randomly extracted,classified and measured under the microscope.Six WT and CD147 overexpressed transgenic mice aged 8 weeks were selected and subjected to 9 rounds of depilation.Hair density was then detected by dermoscopic scanning.The ability of skin wound healing was observed by dorsal drilling,the expression of CD34,a marker gene of skin stem cells,was detected by immunofluorescence staining after frozen section,and the proliferation of skin cells was observed by BrdU incorporation.【Results】Overexpression of CD147 showed no significant effect on the growth and skin structure formation of mice.In compare with wild-type mice,there was no statistical difference in hair length of different types.Classification statistics showed altered proportion of hair types,the proportion of straight hair decreased,and Z-type villi increased.After multiple depilation,CD147 overexpressed mice showed decreased hair density,and increased proliferation of hair follicle stem cells.【Conclusion】CD147 plays an important role in skin hair regeneration.Its expression promotes the proliferation of skin stem cells and accelerates the consumption of hair follicle stem cells after multiple depilations,thus reducing the density of skin hair and the ability of wound healing.
