摘要 目的:探讨非酒精性脂肪性肝纤维化患者病程进展与Slit2-Robol和TGF-β的关系。方法:72例肝纤维化的患者和63例健康志愿者中采集血清,通过RT-qPCR和蛋白质印记分析Slit2、Robo1、TGF-βmRNA和蛋白质表达量;通过酶联免疫吸附实验和免疫组织化学染色检测谷丙转氨酶和谷草转氨酶的含量。通过SPearman法分析患者的血清Slit2、Robo1 mRNA及蛋白表达水平,分析与TGF-β的相关性。结果:与对照组相比,肝纤维化组Slit2、Robo1和TGF-βmRNA和蛋白质表达水平升高,药物治疗组较肝纤维化组Slit2、Robo1和TGF-βmRNA和蛋白质表达水平降低(P<0.05);肝纤维化组谷丙转氨酶和谷草转氨酶(52.17±6.32,72.14±8.68)较对照组(8.34±1.15,15.17±1.38)含量升高(P<0.05),药物治疗组谷丙转氨酶和谷草转氨酶(21.45±2.84,31.73±4.0)较肝纤维化组含量降低(P<0.05)。患者的血清Slit2、Robo1 mRNA及蛋白表达水平均分别与TGF-β呈正相关(P<0.05)。结论:非酒精性脂肪性肝纤维化患者Slit2、Robol和TGF-β水平升高,药物治疗后Slit2、Robol和TGF-β水平降低,推测在肝星状细胞中阻断Slit2-Robo1和TGF-β信号传导可以作为阻断肝纤维化进展的方法。 Objective:To explore the relationship between the progression of non-alcoholic fatty liver fibrosis and Slit2 robol and TGF-β.Methods:Serum samples were collected from 72 patients with liver fibrosis and 63 healthy volunteers.The expression of Slit2,Robo1,TGF-β mRNA and protein were analyzed by RT-qPCR and protein imprinting.The contents of transglutaminase and transglutaminase were detected by enzyme-linked immunosorbent assay and immunohisto chemical staining.The expression levels of serum Slit2,Robo1 mRNA and protein were analyzed by means of spierman method,and the correlation with TGF-β was analyzed.Results:Compared with the control group,the expression levels of Slit2,Robo1,TGF-β mRNA and protein in the liver fibrosis group were higher,and the expression levels of Slit2,Robo1 and TGF-βmRNA and protein in the drug treatment group were lower than those in the liver fibrosis group(P<0.05);in the liver fibrosis group(52.17±6.32,72.14±8.68),compared with the control group(8.34±1.15,15.17±1.38),the content of alanine aminotransferase and glutamic oxaloacetate aminotransferase were higher,and the drug content was higher Compared with the liver fibrosis group,the content of GPT and GST decreased in the treatment group(21.45±2.84,31.73±4.0)(P<0.05).The expression levels of serum Slit2,Robo1 mRNA and protein were positively correlated with TGF-β(P<0.05).Conclusion:The levels of Slit2,robol and TGF-β in patients with nonalcoholic fatty liver fibrosis increased,and the levels of Slit2,robol and TGF-β decreased after drug treatment.It is suggested that blocking the signal transduction of slit2-robo1 and TGF-β in hepatic stellate cells can be used as a method to block the progress of hepatic fibrosis.
