原发性肝细胞癌组织中HIF-1α、RANTES表达水平及与临床病理特征和预后的关系 被引量:6

The expression of HIF-1αand RANTES in primary hepatocellular carcinoma and its relation to clinical pathophysiological characteristics and prognosis

摘要 目的:探究原发性肝细胞癌患者癌组织中缺氧诱导因子1α(hypoxia-inducible factor 1-alpha,HIF-1α)和趋化因子RANTES(regulated on activation,normal T cell expressed and secreted)的表达水平,分析两者与临床病理特征和预后之间的相关性。方法:选取2013年1月至2018年3月于我院就诊的原发性肝癌患者88例。通过免疫组化法(immunohistochemistry,IHC)测定肝癌组织与癌旁组织中HIF-1α、RANTES的表达水平,对比、分析两者与患者临床病理特征和预后情况之间的关系。结果:肝癌组织中HIF-1α、RANTES表达显著高于癌旁组织(P<0.05)。HIF-1α的高表达与分化程度、有无转移相关(P<0.05),而RANTES的高表达则与年龄、分化程度、有无转移等均不相关(P>0.05)。Kaplan-Meier曲线表明,相比于HIF-1α、RANTES低表达组,高表达组患者的术后中位生存时间明显降低(28 vs 48个月,27 vs 43个月,Log Rank P<0.05)。结论:HIF-1α的高表达与癌细胞的增殖、转移相关,同时HIF-1α及RANTES表达水平影响着患者术后生存时间,及时监控两者表达水平有助于有效控制肝细胞癌的发生与发展。 Objective:To explore the expression of hypoxia-inducible factor 1-alpha(HIF-1α)and regulated on activation,normal T cell expressed and secreted(RANTES)in primary hepatocellular carcinoma(HCC),analyze its relationship with clinical pathophysiological characteristics and prognosis.Methods:88 patients with HCC in our hospital from January 2013 to March 2018 were selected.Immunohistochemistry(IHC)was used to detect the expression of HIF-1αand RANTES in observation group(paraffin sections of HCC)and control group(paraffin sections of normal tissue nearby).Furthermore,this was analyzed in combination with clinical pathophysiological characteristic and prognosis to find out their relations.Results:It was showed that the levels of HIF-1αand RANTES in observation group were significantly higher than those in control group(P<0.05).Moreover,the high expression of HIF-1αwas related to differentiation and metastasis(P<0.05),while RANTES had no relationship with age,differentiation and metastasis etc.(P>0.05).Kaplan-Meier curve showed that in comparison with low HIF-1αand RANTES expression patients,the survival time of those who had higher HIF-1αand RANTES expression was much shorter(28 vs 48 months,27 vs 43 months,Log Rank P<0.05).Conclusion:Our data suggest that high expression of HIF-1αis associated with the HCC metastasis.Furthermore,both HIF-1αand RANTES levels are closely related to survival.It is helpful for effectively controlling the development of HCC by monitoring the levels of HIF-1αand RANTES.
