健脾祛痰化瘀方药对脾气虚大鼠心肌细胞线粒体自噬的影响

Effects of Jianpi Qutan Huayu Recipe on mitochondrial autophagy in cardiomyocytes of rats with spleen qi deficiency

目的:观察健脾祛痰化瘀方药对脾气虚大鼠心肌细胞线粒体自噬相关蛋白表达的影响。方法:40只S D大鼠随机分为正常对照组、脾气虚组、中药组以及西药组,每组10只。采用复合法制备脾气虚证模型。中药组给予健脾祛痰化瘀方药,西药组给予阿司匹林和阿伐他汀钙片。Western Blot法检测大鼠心肌AMPK、p-AMPK、ULK1、p-ULK1、LC3-Ⅰ以及L C3-Ⅱ蛋白的表达;利用透射电镜观察大鼠心肌线粒体及线粒体自噬体的超微结构。结果:与正常对照组比较,中药组大鼠心肌p-AMPK/AMPK、p-ULK1/ULK1和LC3-Ⅱ/LC3-Ⅰ显著上调(P﹤0.05,P﹤0.01);与脾气虚组比较,中药组大鼠心肌p-ULK1/ULK1、LC3-Ⅱ/LC3-Ⅰ显著上调(P﹤0.01,P﹤0.05),且均优于西药组(P﹤0.05,P﹤0.01)。脾气虚组大鼠心肌线粒体损伤明显,线粒体数量减少,中药组线粒体损伤有所改善,出现大量的不同阶段的线粒体自噬体。结论:健脾祛痰化瘀方药可能是通过激活AMPK,AMPK直接或者间接提高ULK1磷酸化水平,从而提高大鼠心肌线粒体自噬能力。

Objective:To observe the effects of Gegen Hongteng Jiedu Decoction on toll-like receptor 4(TLR4)and serum interleukin-10(IL-10)in rats with ulcerative colitis(UC),and to analyze the possible mechanism of it.Methods:A total of 60 SPF Wistar male rats were randomly divided into blank group with 10 rats and experimental group.In experimental group,UC animal model was replicated with 2,4,6-trinitrobenzenesulfonic acid(TNBs)/ethanol method,and they were randomly divided into model group,SASP group and Gegen Hongteng Jiedu Decoction group with 15 rats in each one.Give them corresponding durg,and gavage for 14 days.Judging the rats’general conditions,and scored according to disease activity index(DAI),and to observe the changes of TLR4 and IL-10.Results:Compared with blank group,other groups’DAI score and the expression level of TLR4 were increased(P<0.05),while the IL-10 content of serum was decreased(P<0.05).Compared with model group,DAI score and the expression level of TLR4 were decreased(P<0.05)in SASP group and Gegen Hongteng Jiedu Decoction group,while the IL-10 content of serum was increased(P<0.05).There was no statistical significance of Gegen Hongteng Jiedu Decoction group and SASP group.Conclusion:The possible mechanism of Gegen Hongteng Jiedu Decoction may to reduce the expression level of TLR4 and improve the IL-10 content of serum.
