保山猪PRLR、NOCA1和HTRA3基因多态性与其产仔数的关联分析 被引量:7

Correlation of polymorphism of PRLR, NOCA1 and HTRA3 genes with litter size of Baoshan pigs

摘要 采集保山猪耳组织样品共计180头份,记录、统计328窝保山猪经产母猪的总产仔数和产活仔数。通过聚合酶链反应(PCR)、限制性片段长度多态性聚合酶链反应(PCR-RFLP)技术对保山猪的3个繁殖候选基因PRLR、NOCA1、HTRA3的多态性进行研究,分别计算各繁殖基因在保山猪群体中等位基因频率、基因型频率,多态信息含量、卡方值等遗传信息值,并将各繁殖基因多态性和合并基因型的多态性与保山猪产仔数进行关联分析。试验结果表明:3个基因PRLR、NOCA1、HTRA3的各基因型之间产仔数差异不显著(P>0.05),未发现可作为保山猪产仔数候选基因的证据。合并基因型后发现,保山猪繁殖性状PRLR-NOCA1最优分子标记组合为BBBB型,PRLR-HTRA3最优分子标记组合为AAAA型,NOCA1-HTRA3最优分子标记组合为BBAB型。 In this study, a total of 180 ear tissue samples of Baoshan pigs were collected. The total number of 328 nests of born litter size and born alive litter size in the pigs were recorded, counted, and analyzed. The polymorphisms of PRLR, NOCA1, and HTRA3 in the pigs were tested by PCR and PCR-RFLP techniques. The allele frequencies and genotype frequencies, and the polymorphism information content, heterozygosity and chi-square values, of the three productive genes in the Baoshan pigs population, were calculated. The analysis was performed of the association between the polymorphisms of the three productive genes and litter size. The results were as follows: the PRLR, NOCA1 and HTRA3 genes showed no significant difference in litter size among their genotypes(P> 0.05), and no evidence of candidate genes for litter size in the Baoshan pigs was found. The optimal combination of the PRLR-NOCA1 molecular marker was BBBB, the optimal combination of PRLR-HTRA3 molecular marker was AAAA, and the optimal combination of NOCA1-HTRA3 molecular marker was BBAB.
