植物内生真菌来源的倍半萜对LPS诱导细胞炎症反应的抑制作用 被引量:4

摘要 目的:探究马铃薯内生真菌次生代谢产物中新型倍半萜类化合物TA的抗炎作用.方法:脂多糖(LPS)刺激RAW264.7细胞建立体外炎症模型,通过测定细胞存活率、细胞NO的产生量、细胞内活性氧(ROS)水平,细胞培养上清液中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)的释放量、一氧化氮合成酶(iNOS),环氧合酶(COX-2)蛋白表达量,从而评价化合物TA的抗炎活性.结果:化合物TA对细胞无毒性,可剂量依赖性地抑制LPS刺激产生的NO,降低ROS水平,减少细胞炎症因子TNF-α和IL-1β的产生及下调iNOS,COX-2蛋白的表达量.结论:马铃薯内生真菌次生代谢产物中新型倍半萜类化合物TA具有显著的抗炎活性. Objective:To investigate the anti-inflammatory effect of a new sesquiterpene compound TA in the secondary metabolites of potato endophytic fungi.Methods:Lipopolysaccharide(LPS)stimulated RAW264.7 cells to establish an in vitro inflammation model.Cell viability,nitrite levels representing NO production,intracellular reactive oxygen species(ROS)levels,the release of inflammatory cytokines into the supernatant such as tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)were measured.Moreover,Western-blotting assays were used to detect the expression of induced-nitric oxide synthase(iNOS)and cyclooxygenase-2(COX-2)proteins to evaluate the anti-inflammatory activity of compound TA.Results:Compound TA was non-toxic to cells,and could inhibit NO produced,reduce ROS level,decrease the production of inflammatory cytokines and down-regulate the expression of iNOS and COX-2 proteins in a dose-dependent manner.Conclusion:TA,a novel sesquiterpene compound in the secondary metabolites of potato endophytic fungi,has significant anti-inflammatory activity.
