摘要
目的:探讨二至丸对去卵巢所致AD模型小鼠学习记忆的影响。方法:小鼠去卵巢后6周开始给药,假手术组与模型组灌胃等容量的蒸馏水,二至丸高、低剂量组分别灌胃二至丸混悬液2g生药/kg、1g生药/kg,阳性对照组隔天皮下注射苯甲酸雌二醇0.02mg,每日1次,连续32d。给药20d后八臂迷宫,Y迷宫检测小鼠学习能力。HE染色和尼氏染色观察海马形态学,免疫组化法检测海马Aβ含量。结果:与假手术组相比,模型组小鼠在Y迷宫中正确交替率明显降低;在八臂迷宫进入有效臂所需的总时间、总错误次数明显上升,首错前正确数降低;与模型组比较,二至丸给药组小鼠正确交替率明显提高,进入有效臂所需的总时间与总错误次数明显下降,首错前正确升高。二至丸可增加海马CA1区细胞总数,减少坏死细胞数;减少海马CA1区Aβ阳性细胞数。结论:二至丸能够改善去卵巢AD小鼠的学习记忆能力,可能与其神经元保护作用和减少Aβ沉积有关。
Objective:To investigate the effects of erzhi pill on learning and memory in Alzheimer's disease(AD)mice induced by ovariectomy.Methods:After 6 weeks of operation,the mice in the sham operation group and the model group were given the same volume of distilled water,while Erzhi pill high and the low dose group were given the drug suspension 2g crude drug/kg,1g crude drug/kg,positive control group mice were injected with estradiol benzoate 0.02 mg twice a day for 32 days.After 20 days of drug administration,the learning ability of mice was measured by eight-arm labyrinthine and Y maze.The hippocampal morphology was observed by HE staining and Nissl staining after the behavioral test.The content of Aβin hippocampus was detected by immunohistochemistry.Results:Compared with sham operation group,model group mice in the Y maze test right alternation rate decreased significantly;in the eight arm maze testthe total time required for the mice to enter the effective arm andthe whole wrong times were significantly increased,the first wrong before the correct number is reduced.Compared with the model group,Erzhi pill treatment group mice right alternating rate enter the total time significantly improved,effective arm required and total number of errors decreased significantly,the first wrong before the correct number rise.Erzhi pill can increase the total number of CA1 cells in hippocampus,reduce the number of necrotic cells,and decrease the number of Aβpositive cells in the hippocampus CA1 region.Conclusion:Erzhi pill can improve the learning and memory ability of ovariectomized AD mice,which may be related to the protective effect of neurons and the reduction of Aβdeposition.
