玉米干种子DNA快速提取技术研究

Study on Rapid Extraction of DNA from Maize Seeds

摘要 以玉米干种子为提取材料,采用磁性纳米微球作为吸附介质进行基因组DNA的提取,分别探讨不同裂解液、裂解温度、时间和磁珠用量对基因组DNA提取效率的影响。结果表明裂解液2%CTAB、裂解温度65℃、裂解时间10 min、磁珠用量15μL时基因组DNA提取浓度高、纯度好、适宜后续实验。 Using maize seeds as materials, extracted the genomic DNA by magnetic nanospheres.The results showed that when the lysate was 2%CTAB, the pyrolysis temperature was 65 ℃, the time was 10 min and the dosage of magnetic beads was 15 μL, the genomic DNA extraction concentration was high, the purity was good, and it was suitable for subsequent experiments.
