黑色素瘤缺乏因子2在胆管癌组织中的表达及其临床意义 被引量:3

摘要 目的:研究黑色素瘤缺乏因子2(AIM2)在胆管癌组织中的表达,并分析其表达水平与胆管癌患者临床病理特征及预后的关系。方法:采用组织芯片技术和免疫组织化学染色法,分别检测91例胆管癌组织及其部分正常组织中AIM2的表达水平,采用Wilcoxon秩和检验比较癌组织及正常组织中AIM2表达水平的差异,采用χ^2检验分析胆管癌组织中AIM2表达水平与患者临床病理特征的关系,采用Kaplan-Meier法分析胆管癌组织中AIM2表达水平与患者总生存期的关系,拟合Cox模型评价不同指标的预后价值。结果:AIM2在正常组织中高表达(P<0.005),而在胆管癌组织中以不表达和低表达为主;AIM2的表达水平与肿瘤大小显著相关(χ^2=7.380,P=0.007);AIM2低表达的胆管癌患者总生存期(OS)较AIM2高表达水平的患者显著缩短(HR=0.293,95%CI:0.130~0.662,P=0.002);多因素Cox风险比例模型显示,AIM2表达水平(HR=0.320,95%CI:0.120~0.853,P=0.023)、性别(HR=2.268,95%CI:1.019~5.044,P=0.041)和年龄(HR=0.180,95%CI:0.070~0.465,P=0.000)均可作为胆管癌患者预后评判的独立风险因素。结论:AIM2在胆管癌组织中呈低表达,有望成为胆管癌患者的预后预测指标。 Objective:To investigate the expression of absent in melanoma 2(AIM2) in cholangiocarcinoma tissues,and its correlation with patients′ clinico-pathological characteristics and prognosis.Methods:Tissue microarray and immunohistochemistry was used to examine the AIM2 expression in 91 cases of cholangiocarcinoma tissues and some of its normal tissue.Wilcoxon rank sum test was used to compare the expression of AIM2 in cholangiocarcinoma and corresponding normal tissues.The chi-square test was used to analyze the relationship between the AIM2 expression in cholangiocarcinoma tissues and clinicopathological features of the patients.Kaplan-Meier survival analysis was performed to analyze the correlation between AIM2 expression and patients′ overall survival,and the Cox model was used to analyze the correlation between different clinical parameters and prognosis.Results:The immunohistochemistry results showed that AIM2 was highly expressed in normal tissues(P<0.005),with no or low expression in cholangiocarcinoma tissues.The expression level of AIM2 was significantly correlated with tumor size(χ^2=7.380,P=0.007).The overall survival(OS) of patients with low expression of AIM2 was significantly shorter than that of patients with high AIM2 expression(HR=0.293,95%CI:0.130-0.662,P=0.002).Multi-factor Cox model analysis indicated that the expression level of AIM2(HR=0.320,95%CI:0.120-0.853,P=0.023) and gender(HR=2.268,95%CI:1.019-5.044,P=0.041)and age(HR=0.180,95%CI:0.070-0.465,P=0.000)were independent prognostic factors of cholangiocarcinoma patients.Conclusion:AIM2 is expressed lowly in cholangiocarcinoma tissue,and it is expected to be an important prognostic indicator for the cholangiocarcinoma patients.
