摘要
目的:探讨热休克蛋白A12B (HSPA12B)对β淀粉样蛋白(Aβ)诱导的PC12细胞tau蛋白磷酸化的影响。方法:将处于对数生长期的PC12细胞分为4组:对照组、DMSO组、Aβ(20μmol/L)刺激组、Aβ刺激HSPA12B-siRNA组(Aβ+HSPA12B-siRNA)。采用Western blot法检测4组细胞的HSPA12B、磷酸化tau蛋白(p-tau)表达水平。结果:4组PC12细胞HSPA12B、p-tau蛋白表达水平比较差异均有统计学意义(P<0.05);与对照组比较,Aβ组PC12细胞HSPA12B、p-tau蛋白表达均上调(P>0.05),DMSO组无显著差异(P>0.05);而与Aβ组比较,Aβ+HSPA12B-siRNA组PC12细胞HSPA12B下调、p-tau蛋白表达均增强(均P<0.05)。结论:HSPA12B调节Aβ诱导的PC12细胞tau蛋白磷酸化,进而影响阿尔茨海默病的病程。
Objective:To explore the effect of heat shock proteins A12 B(HSPA12 B) on tau hyperphosphorylation in Aβ-induced PC12 cells.Methods: PC12 cells in logarithmic growth phase were divided into four groups:the normal group,DMSO group,Aβ group(stimulated by Aβ 20 μmol/L),and the Aβ+HSPA12 B-siRNA group.Western blot was used to detect the expression of HSPA12 B and tau phosphorylation(p-tau) in PC12 cells of the four groups.Results: There were significant statistically differences in the expression of HSPA12 B and p-tau protein in PC12 cells among the four groups(P<0.05).Compared with normal group,the expression of HSPA12 B,p-tau protein in PC12 cells was up-regulated in Aβ group(P<0.05),while there′s no difference in DMSO group(P>0.05).By knocking down the expression of HSPA12 B,Aβ-induced tau hyperphosphorylation in PC12 cells were significantly increased than that of Aβ group(P<0.05).Conclusion: HSPA12 B regulates the phosphorylation of tau protein in PC12 cells induced by Aβ,and then affects the course of Alzheimer′s disease.
