玉木耳总黄酮的提取工艺及抗氧化活性分析 被引量:4

Extraction Technology of Total Flavonoids from Auricularia cornea var.Li.and Analysis of Its Antioxidant Activity

摘要 【目的】优化玉木耳(Auricularia cornea var.Li.)总黄酮的提取工艺,并对玉木耳、黑木耳(Auricularia auricula)和毛木耳(Auricularia polytricha)总黄酮的抗氧化活性进行比较。【方法】采用超声波破碎乙醇溶液浸提法提取玉木耳总黄酮,通过单因素实验和正交实验优化提取工艺,并采用ABTS法、水杨酸法、邻苯三酚自氧化法和DPPH法对3种木耳总黄酮的抗氧化活性进行测定。【结果】玉木耳总黄酮提取工艺的最佳参数为:液固比15mL∶1g、乙醇体积分数80%、超声波破碎功率80W、浸提时间8h,在这一参数下得到玉木耳、毛木耳和黑木耳的总黄酮提取量分别为3.44,8.78,7.02mg·g^-1。玉木耳的总抗氧化活性以及对羟基自由基、超氧阴离子和DPPH的清除能力均强于其他两种木耳。【结论】研究结果为玉木耳天然有效成分的研究和开发提供了参考资料。 [Purposes]Optimized the extraction process of Auricularia cornea var.Li,and compared the antioxidant activities of A.cornea,Auricularia auricula and Auricularia polytrichaflavonoids.[Methods]The total flavonoids of A.cornea were extracted by the method of ultrasonic breaking ethanol solution extraction.The extraction process was optimized by single factor experiment and orthogonal experiment.The antioxidant activities of the three kinds of total flavonoids were determined by ABTS,salicylic acid,pyrogallol autoxidation and DPPH.[Findings]The best extraction parameters of total flavonoids fromA.cornea were:liquid-solid ratio of 15mL∶1g,ethanol volume fraction of 80%,ultrasonic power of 80W,extraction time of 8h.under these parameters,the total flavonoids extracted fromA.cornea,A.polytricha,and A.auricula were 3.44,8.78,7.02mg·g^-1,respectively.The total antioxidant activity and scavenging capacity of hydroxyl radicals,superoxide anion and DPPH of A.cornea were higher than those of the other two species.[Conclusions]The results provide a reference for the research and development of the natural active components of A.cornea.
