基于代谢组学的陈皮藿香汤及其单味药对腹泻大鼠的干预作用研究 被引量:10

Study on the Intervention Effect of Chenpi Huoxiang Decoction and Its Single Herb on Diarrhea Rats Based on Metabonomics

摘要 目的研究陈皮藿香汤及其单味药对腹泻大鼠血清代谢组的影响,并初步探讨其作用通路。方法采用番泻叶煎剂(0.5 g·mL^-1,10 mL·kg^-1)灌胃14 d(每天2次)建立大鼠腹泻模型。将SD大鼠随机分为对照组、模型组、陈皮藿香汤组(0.5 g·mL^-1,1 mL·kg^-1)、陈皮组(0.25 g·mL^-1,1 mL·kg^-1)、广藿香组(0.25 g·mL^-1,1 mL·kg^-1),每组10只;灌胃给药,每天1次,连续7 d。采用核磁共振氢谱(1H-NMR)技术检测大鼠血清,以主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)法筛选出相关差异代谢物,并对其代谢通路进行分析。结果 PCA得分图显示,对照组与模型组有明显的分离趋势,3个给药组的样本点均有向对照组靠近的趋势。OPLS-DA分析显示,与对照组比较,模型组的血清样本中分别发现了16个差异代谢物(VIP> 1,P <0.05);与模型组比较,陈皮藿香汤组、陈皮组和广藿香组的血清样本中分别发现了11、12和10个差异代谢物(VIP> 1,P <0.05),且3个给药组中差异代谢物的变化趋势基本与对照组的变化趋势一致。差异代谢物主要与丁酸代谢以及酮体的合成和降解通路有关。结论陈皮藿香汤及其单味药能够改善腹泻模型大鼠血液代谢,可能主要与丁酸代谢以及酮体代谢有关。 Objective The effects of Chenpi Huoxiang decoction and its single herb on serum metabolomics of diarrhea rats were studied and the pathways were preliminarily explored. Methods The rat diarrhea model was established by intragastric administration of Sennae folium decoction(0.5 g·mL^-1,10 mL·kg^-1)for 14 days(twice a day). The rats were randomly divided into control group,model group,Chenpi Huoxiang decoction group(0.5 g·mL^-1,1 mL·kg^-1),Citri reticulatae pericarpium decoction group(0.25 g·mL^-1,1 mL·kg^-1)and Pogostemonis herba single decoction group(0.25 g·mL^-1,1 mL·kg^-1),with 10 rats in each group,and the drug was given by intragastric administration once a day for 7 days. The rats’ serum were detected by nuclear magnetic resonance hydrogen spectroscopy(1H-NMR). The related differential metabolites were screened by principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA),then the metabolic pathways were analyzed.Results PCA score plots showed that there was an obvious trend of separation between the control group and the model group, and the sample points of the three administration groups all tended to approach the control group.OPLS-DA analysis showed that, compared with the control group, 16 differential metabolites were found in the serum samples of the model group(VIP > 1,P < 0.05). Compared with the model group,11,12 and 10 differential metabolites were found in the serum samples of the Chenpi Huoxiang decoction group,Citri reticulatae pericarpium group and Pogostemonis herba group respectively(VIP > 1, P < 0.05), and the changing of the differential metabolites in the three administration groups was basically consistent with that of the control group. The differential metabolites were mainly related to butyric acid metabolism and ketone body synthesis and degradation pathways.Conclusion Chenpi Huoxiang decoction and its singlet herbs can improve the blood metabolism of diarrhea rats,which may be mainly related to butyric acid metabolism and ketone metabolism.
