高效液相色谱法测定体外透皮样品中氧化苦参碱的含量 被引量:1

摘要 目的:建立体外透皮样品中氧化苦参碱含量的测定方法。方法:采用高效液相色谱法测定体外透皮样品,Spherisorb ODS2 C18色谱柱(4.6 mm×200 mm,5μm),流动相为乙腈-0.3%磷酸水溶液(10∶90,V∶V),并用三乙胺调节pH至3.1,流速为1.0 ml/min,检测波长为207 nm,柱温为60℃。结果:氧化苦参碱在1.055~42.2μg/ml范围内,呈现良好的线性关系(R2=0.9999)。氧化苦参碱体外透皮样品在24 h内稳定性良好,方法的精密度良好,低、中、高3种浓度的平均回收率依次为(99.7±0.6)%、(100.2±0.5)%、(100.6±0.4)%,符合要求。结论:所建高效液相色谱法可用于测定体外透皮样品中氧化苦参碱的含量。 Objective:To determinate the concentration of oxymatrine in transdermal receptor solution.Methods:HPLC method was established and a Spherisorb ODS2 C18 column(4.6 mm×200 mm,5μm)was adopted in this study.The mobile phase was consisted of acetonitrile-0.3%phosphoric acid water solution(10∶90,V∶V)and the pH was adjusted to 3.1 by triethylamine.The flow rate was 1.0 ml/min and the column temperature was set as 60℃.The detection wavelength was set at 207 nm.Results:The content of oxymatrine had a good linear relationship in range of 1.055-42.2μg/ml(R^2=0.9999).The test solution was stable in 24 h and the method was of good precision.The mean recoveries of oxymatrine in low,medium,and high concentration solution was(99.7±0.58)%,(100.2±0.51)%and(100.6±0.43)%,respectively.Conclusion:The HPLC method can be applied to determine the concentration of oxymatrine in transdermal receptor solution.
