过度瘦身对雌性大鼠血清性激素的影响及左归丸的干预效应

The Effect of Excessive Weight Loss on Serum Sex Hormone of Female Rats and the Intervention Effect of Zuogui Pill(左归丸)

摘要 目的:探讨运用"左旋肉碱+半量饮食+过度运动"方法造成大鼠过度瘦身状态后,对其生殖功能的影响以及给予左归丸的干预效应。方法:选取动情周期规律的雌鼠,随机分为造模4周组(A组)、造模8周组(B组)两大组,每组依次分为正常组、模型组、恢复饮食组、治疗组。造模结束后,运用酶联免疫吸附试验(ELISA)法检测血清中雌二醇(E2)、卵泡刺激素(FSH)、黄体生成激素(LH)的表达水平。结果:(1)体质量:与正常组比较,造模4周及8周大鼠体质量下降明显,差异有统计学意义(P<0.01)。且造模4周体质量下降17%,8周体质量下降23%。(2)动情周期:与正常组比较,造模4周及8周大鼠动情周期紊乱,差异有统计学意义(P<0.01)。(3)血清性激素检测:与正常组比较,造模4周及8周大鼠FSH、LH水平均升高,E2水平下降,差异有统计学意义(P<0.01);造模4周及8周后,恢复饮食组FSH、LH均低于模型组,E2水平高于模型组,差异有统计学意义(P<0.01);造模4周,治疗组FSH、LH均低于恢复饮食组,E2水平高于恢复饮食组,差异均有统计学意义(P<0.01或P<0.05)。其余各组干预前后差异无统计学意义(P>0.05)。结论:通过"左旋肉碱+半量饮食+过度运动"方法可造成大鼠过度瘦身状态;当体质量下降17%时,大鼠动情周期明显紊乱,FSH、LH升高及E2下降明显;当体质量下降23%时,变化更为明显。左归丸可在一定程度上改善大鼠动情周期及体质量变化,可有效上调E2及下调LH、FSH水平,但在瘦身8周后,恢复效果较差。 Objective:To explore the effect of"L-carnitine+half-dose diet+excessive exercise"on the reproductive function of rats after excessive weight loss and the intervention effect after Zuogui pill.Methods:Female rats with regular estrous cycle were selected and randomly divided into two groups:4 weeks modeling group(group A)and 8 weeks modeling group(group B).Each group was divided into normal group,model group,diet recovery group,and treatment group.After the modeling was completed,the expression levels of estradiol(E2),follicle stimulating hormone(FSH)and luteinizing hormone(LH)in serum were detected by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the normal group,the body weight of rats decreased significantly at 4 and 8 weeks after modeling,and the difference were statistically significant(P<0.01).The body weight decreased by 17%after 4 weeks of modeling,and decreased by 23%after 8 weeks of modeling.Compared with the normal group,the estrous cycle of rats was disordered at 4 and 8 weeks after modeling,and the difference were statistically significant(P<0.01).Compared with the normal group,the FSH and LH levels of the rats were increased and the E2 level was decreased in the 4 and 8 weeks after modeling(P<0.01).After 4 weeks and 8 weeks of modeling,the FSH and LH levels of the recovery diet group were lower than the model group,and the E2 level was higher than the model group,and the difference were statistically significant(P<0.01).After 4 weeks of modeling,the FSH and LH levels of the treatment group were lower than the recovery diet group,and the E2 level was higher than the recovery diet group,and the differences were statistically significant(P<0.01 or P<0.05).There was no significant difference between the other groups before and after intervention(P>0.05).Conclusions:The method of"L-carnitine+half-dose diet+excessive exercise"can cause excessive weight loss in rats.When the body weight decreased by 17%,the estrous cycle of the rats was obviously disordered,the levels of FSH,LH were significantly increased,and the level of E2 was significantly decreased.When the body weight decreased by 23%,the change was more obvious.Zuogui pill can improve the estrous cycle and body weight changes of rats to a certain extent,and can effectively increase E2 and reduce LH and FSH levels,but after losing weight for 8 weeks,the recovery effect is poor.
