苓桂术甘汤对瘦素缺陷肥胖ob/ob小鼠卵巢SR-B1蛋白表达影响

Effects of Linggui Zhugan Decoction on the Expression of SR-B1 Protein in Ovary of Leptin-deficient Obese Mice

目的观察苓桂术甘汤对瘦素缺陷肥胖ob/ob小鼠生殖功能的影响,探讨其作用机制是否与其干预卵巢SR-B1蛋白有关。方法选用雌性肥胖ob/ob小鼠24只,雌性野生型(WT)小鼠8只,实验分为WT组、ob/ob组、苓桂术甘汤大剂量组(LGZGH)、苓桂术甘汤小剂量组(LGZGL),灌胃给药4周。观察子宫和卵巢形态,称取子宫重量,计算子宫指数;采用HE染色法检测卵巢组织形态学改变;采用蛋白质免疫印迹法(Western blot)检测卵巢组织SR-B1蛋白表达。结果与WT组比较,ob/ob组小鼠子宫管腔狭窄,管壁变薄,弹性差;子宫重量和子宫重量指数减少(P<0.01),卵巢组织结构破坏,可见卵泡闭锁,卵巢组织SR-B1蛋白表达降低;与ob/ob组比较,苓桂术甘汤小剂量组可以增加子宫重量和子宫重量指数(P<0.01),苓桂术甘汤可以减轻卵巢组织结构的破坏,显著上调卵巢组织SR-B1蛋白表达。结论苓桂术甘汤可以部分改善瘦素缺陷肥胖ob/ob小鼠生殖功能障碍,作用机制可能与其调控卵巢组织SR-B1蛋白表达有关。

Objective To observe the effect of Linggui Zhugan Decoction on reproductive function of leptin-deficient(ob/ob)mice and to investigate whether its mechanism was related to the intervention of SR-B1 protein in ovary.Methods Twenty-four obese female ob/ob mice and eight female wild-type(WT)mice were used.The experiment was divided into WT group,ob/ob group,large dose group of Linggui Zhugan Decoction(LGZGH)and low dose group of Linggui Zhugan Decoction(LGZGL),continuously intragastric administration.After 4 weeks,the shape of uterus and ovary were observed.The uterine index were calculated.Ovarian histomorphology was detected by HE staining.SR-B1 expressions in ovary were detected by Western blot.Results Compared with WT group,the uterine in ob/ob group was narrow.The uterine weight index was decreased(P<0.01).The structure of ovary was destroyed.The expression of SR-B1 protein in ovarian was increased.Compared with ob/ob group,low dose group of Linggui Zhugan Decoction could increase uterine weight index(P<0.01).It could alleviate the destruction of ovariantissue structure.The expression of SR-B1 protein in ovarian tissue was up-regulated.Conclusion Linggui Zhugan Decoction can partially improve the reproductive dysfunction of leptin-deficient mice,and the mechanism may be related to the regulation of SR-B1 protein expression in ovarian tissues.
