新安名方脑络欣通HPLC指纹图谱研究

摘要 目的运用高效液相色谱技术和主成分分析思想建立并分析脑络欣通指纹图谱。方法HPLC法建立脑络欣通指纹图谱,色谱柱:Unitary C18(4.6 mm×250 mm,5μm),流动相:甲醇(A)-水(B),检测波长:270 nm,流速:0.8 mL/min,柱温:27℃,进样量:20μL,时间梯度洗脱,分析时间:110 min,并通过标准品比对指认化学成分;相似度分析法分析12批样品相似度;主成分分析法分析12批样品主成分分布情况。结果初步建立了脑络欣通HPLC指纹图谱,标记了11个特征共有峰;经标准品比对指认了其中4个成分,分别是对羟基苯甲醇、紫丁香苷、毛蕊异黄酮苷、芒柄花苷;12批样品相似度在0.872~0.996;主成分分析指认出4个主成分,累积贡献率为80.01%,其中第7批样品综合得分排名第一。结论该实验建立了脑络欣通HPLC指纹图谱,指认了4个特征共有峰,12批样品相似度较好,且第7批组方药材综合评价第一。该部分研究是为了能更好的提高临床用药安全性,优化剂型,并为下一步谱效关系研究提供基础。 Objective To establish and analysis the HPLC fingerprint of Naoluo Xintong Decoction by using the principle of high performance liquid chromatography and principal component analysis.Methods Fingerprint of Naoluo Xintong was established by HPLC,the analysis was performed on the Unitary C18(4.6 mm×250 mm,5μm)column with a mobile phase consisting of Methanol(A)-Water(B)with gradient elution;the detection wavelength was 270 nm;the flow rate was 0.8 mL/min;the column temperature was 27℃;the injection volume was 20μL and the analysis time was 110 min,and the components were identified by standard comparison;the similarity analysis method was used to analyze the similarity of 12 batches of samples;using principal component analysis method to analyze the distribution of principal components score of 12 batches of samples.Results The HPLC fingerprint of Naoluo Xintong Decoction was preliminarily established,and 11 characteristic common peaks were marked,and four constituents(4-hydroxybenzyl alcohol,syringin,calycosin-7-O-glucoside,ononin)were identified by standard comparison.The similarities of samples were between 0.872-0.996;Principal component analysis identifies four principal components,the cumulative contribution rate was 80.01%,and the seventh batch of samples ranked first in comprehensive score.Conclusion In this study,HPLC fingerprint of Naoluo Xintong was established,identified four characteristic common peaks,the similarity of the 12 batches of samples was good and the comprehensive evaluation of the 7 th batch of prescription was the first,which can be used for improving the safety of clinical medication,optimizing the dosage form,and providing the basis for the next study of spectral relationship.
