基于数据挖掘和整合药理学平台牛膝芍药治疗骨关节炎成分-靶标-通路分析 被引量:3

Analysis on“Component-target-pathway”of Achyranthes Bidentata Blume and Paeonia Tacti Lora Pall Drug Pair in Treating Osteoarthritis Based on Mining and Integrated Pharmacology

摘要 目的以牛膝-白芍药对为研究对象,借助整合药理学平台(www.tcmip.cn)探究其对骨关节炎的作用机制。方法利用整合药理学平台将牛膝-白芍的化学成分进行靶标预测,与关节炎(osteoarthritis,OA)相关疾病靶标信息进行蛋白-蛋白相互作用(PPI)网络构建,富集分析该药对治疗OA的关键靶标,构建牛膝-白芍药对化学成分、关键作用靶标和疾病相关通路的相互作用网络,绘制"中药-化学成分-关键靶标-通路"的可视化多维网络,结合相关文献报道对牛膝-白芍药对治疗OA的作用机制进行预测。结果预测牛膝化学成分15种,包括多糖类、皂苷类、甾酮类、黄酮类等。预测白芍化学成分50种,包括苷类、萜类、黄酮类、鞣质类等。其中药物和疾病的共同作用靶标PRKAA1、PRKAA2、PRKAB1、NF-KB1、NF-KB2、NF-KB1A、CXCR1、BCL2、TP53。其中疾病通路包括内分泌系统、前列腺癌、乙型肝炎、甲状腺激素、癌症、循环系统、神经系统、神经营养因子、细胞生长和死亡、FoxO信号通路等。结论在整合药理学平台的基础上,该研究初步揭示了牛膝-白芍药对治疗OA的潜在活性成分和可能作用机制,为进一步探究牛膝-白芍治疗骨关节炎作用机制提供了理论支持和指导。 Objective Taking Achyranthes bidentata Blume and Paeonia tacti lora Pall drug pair as research object,to explore its mechanism of action on osteoarthritis with integrated pharmacological platform(www.tcmip.cn).Methods The integrated pharmacological platform was used to predict the chemical constituents of Achyranthes bidentata Blume and Paeonia tacti lora Pall drug pair,and a protein-protein interaction(PPI)network was constructed with target information of osteoarthritis(OA)related diseases.Key targets for treating OA,construct an interaction network of Achyranthes bidentata Blume and Paeonia tacti lora Pall drug pair on chemical composition,key role targets and disease-related pathways,draw a visual multi-dimensional network of"Chinese medicine-chemical composition-key targets-pathways",combined with relevant literature reports,the mechanism of the effect of Achyranthes bidentata Blume and Paeonia tacti lora Pall drug pair on OA was predicted.Results The Achyranthes bidentata Blume chemical composition 15 was predicted,including polysaccharides,saponins,anthrones,flavonoids and the like.It was predicted that 50 kinds of chemical constituents of Paeonia tacti lora Pall,including glycosides,anthraquinones,flavonoids and tannins.Among them,drugs and diseases had a common target of PRKAA1,PRKAA2,PRKAB1,NF-KB1,NF-KB2,NF-KB1 A,CXCR1,BCL2,and TP53.The disease pathway includes endocrine system,prostate cancer,hepatitis B,thyroid hormone signaling pathway,cancer pathway,circulatory system,nervous system,neurotrophin signaling pathway,cell growth and death,and FoxO signaling pathway.Conclusion Based on the integration of the pharmacological platform,this study has revealed the potential active ingredients and possible mechanism of Achyranthes bidentata Blume and Paeonia tacti lora Pall drug pair medicine for treating OA,which provides theoretical support and guidance for further exploring the mechanism of action of Achyranthes bidentata Blume and Paeonia tacti lora Pall drug pair on osteoarthritis.
