摘要
以灵宝苹果榨汁后所得果渣为原料,利用纤维素酶提取果渣中的果胶,通过单因素试验分别研究了酶浓度、料液比、pH值、提取时间和提取温度对果胶提取率的影响。以果胶产率为评价指标,选取不同因素及水平,设计L9(34)正交试验,优化得到最佳提取试验条件,即果渣料液比为1:15(g/mL),浸提溶液pH为4.5,酶解反应温度为50℃,纤维素酶浓度为0.3%,浸提时长4h。在最佳试验条件下果胶提取率达到11.0%,该方法操作简单,重现性好。
The Ling-bao Apple pomace was used as experimental materials by the cellulose.By single factor experiment studied the effect of temperature,ratio of material to liquid,pH value,extraction time.Then taking pectin yield as the evaluation index,A L9(34)orthogonal test was designed,and the best extraction conditions were optimized,the result showed that:the ratio of fruit residue to liquid was 1:15(g/ml),the pH of extraction solution was 4.5,the temperature of enzymatic hydrolysis reaction was 50℃,the concentration of cellulase was 0.3%,and the extraction time was 4 hours.At the lost,the pectin extraction rate was 11.0%under the optimum conditions.The method was simple and reproducible.
