黄芪提取物对心肌梗死大鼠血清cTnI、TGF-β1的影响 被引量:3

摘要 目的探究黄芪提取物对心肌梗死大鼠血清心肌肌钙蛋白I(cardiac Troponin I,cTnI)、转化生长因子-β1(Transforming growth factor-β1,TGF-β1)水平的影响。方法选取30只SD健康雄性大鼠,随机选取10只大鼠作为正常组,对剩余20只大鼠进行心肌梗死模型建立,并分为心肌梗死组、黄芪干预组2组。对3组大鼠心功能指标进行检测,酶联免疫吸附试验法检测cTnI、TGF-β1水平,Western blot法检测磷酸化蛋白激酶(p-AKT)、Bax蛋白和B淋巴细胞瘤-2基因(B-cell lymphoma-2,Bcl-2)相对表达量。结果黄芪干预组大鼠各项心功能指标水平均优于正常组,差异有统计学意义(P<0.05)。心肌梗死组大鼠心肌组织cTnI、TGF-β1水平均高于正常组,差异有统计学意义(P<0.05);黄芪干预组大鼠心肌组织cTnI、TGF-β1水平均高于正常组,低于心肌梗死组,差异有统计学意义(P<0.05)。心肌梗死组大鼠细胞凋亡相关蛋白p-AKT、Bcl-2相对表达量均显著高于正常组,Bax相对表达量低于正常组,差异有统计学意义(P<0.05);黄芪干预组大鼠细胞凋亡相关蛋白p-AKT、Bcl-2相对表达量均显著高于正常组,低于心肌梗死组,Bax相对表达量低于正常组,高于心肌梗死组,差异有统计学意义(P<0.05)。结论黄芪提取物能够有效改善心肌梗死模型大鼠的心功能,抑制心肌细胞凋亡,为心肌梗死症状的临床治疗提供了理论帮助。 Objective To explore the effects of astragalus extract on the levels of serum cardiac Troponin I(cTnI)and transforming growth factor-β1(TGF-β1)in rats with myocardial infarction.Methods Thirty healthy SD male rats were enrolled in the study.The 10 rats were enrolled in control group,and the myocardial infarction models were established in the other 20 rats,then the rats were divided into myocardial infarction group and astragalus intervention group.The cardiac function indexes were detected.Moreover,the levels of cTnI and TGF-β1 were detected by ELISA,and the relative expression levels of phosphorylated protein kinase(PAKT),Bax protein and b-cell lymphoma-2(Bcl2)were detected by Western Blot.Results The heart function indexes in astragalus intervention group were superiot to those in control group(P<0.05).The levels of cTnI and TGF-β1 and the relative expressions levels of apoptosis-related proteins PAKT and Bcl2 in myocardial tissue of rats in astragalus intervention group were significantly higher than those in control group,however,which were significantly lower than those in myocardial infarction group(P<0.05).Moreover,the relative expressions levels of apoptosis-related proteins PAKT and Bcl2 and the levels of cTnI and TGF-β1 in myocardial infarction group were significantly higher than those in control group,but the relative expression levels of Bax were significantly lower than those in control group(P<0.05).And the relative expression levels of Bax were significantly lower than those in control group,however,which were significantly higher than those in myocardial infarction group(P<0.05).Conclusion Astragalus extract can effectively improve the cardiac function of the rats of myocardial infarction model,inhibit the apoptosis of myocardial cells and provide theoretical help for clinical treatment.
