摘要
目的观察芒果苷对自发性高血压大鼠外周血、肾脏T淋巴细胞亚群和血清炎性因子的影响,探讨芒果苷对高血压T淋巴细胞亚群和炎症相关因子的变化以及细胞免疫调节的作用机制,为芒果苷在抗炎免疫领域的应用提供新思路。方法以40只雄性自发性高血压大鼠随机分为模型组、苯那普利组、芒果苷低剂量给药组、芒果苷中剂量给药组、芒果苷高剂量给药组,以8只Wistar Kyoto大鼠作为空白对照组。给药8周后,采用流式细胞术测定外周血和肾脏CD4+、CD8+细胞数量百分比;采用ELISA法测定血清中白细胞介素(IL)-6、IL-10、TNF-α和肾匀浆液中IL-4、IFN-γ的含量。结果与模型组比较,芒果苷各剂量组外周血CD4+/CD8+比值均显著降低(P<0.01),芒果苷中、高剂量与模型组比较,肾脏组织CD4+/CD8+比值均显著降低(P<0.01)。与模型组比较,芒果苷可显著降低大鼠血清IL-6、TNF-α和肾组织IL-4的含量(P<0.05或P<0.01)。芒果苷中、高剂量与模型组比较,可显著升大鼠血清IL-10和肾组织IFN-γ的含量(P<0.05或P<0.01)。结论结果表明高血压伴随着炎症反应,芒果苷具有明显的的抗炎作用,其机制可能与调控T淋巴细胞亚群比例,减少促炎因子的产生,从而减轻高血压的症状及靶器官损伤。
Objective To observe the effects of Mangiferin on peripheral blood,renal T lymphocyte subsets and serum inflammatory factors in spontaneously hypertensive rats and investigatethe changes of Mangiferin on T lymphocyte subsets and inflammatory related factors in hypertension and the mechanism of cellular immune regulation.The new ideas for the application of mangiferin in the field of anti-inflammatory and immune system were provided.Methods Forty male spontaneously hypertensive rats were randomly divided into a model group,a benazepril group,a low dose mangiferin group,a middel dose mangiferin group,a high dose mangiferin group,and 8 Wistar Kyoto rats as a blank control group.Results After 8 weeks administration,the numbers of CD4+and CD8+cells in peripheral blood and kidney were measured by flow cytometry,and the contents of IL-4,IL-6,IL-10,TNF-αand IFN-γin kidney homogenate were measured by ELISA.Compared with the model group,the ratio of CD4+/CD8+in the peripheral blood of mangiferin groups decreased significantly(P<0.01).Compared with the model group,the ratio of CD4+/CD8+in renal tissue in middle and high dose mangiferin groups was significantly lower than that in the model group(P<0.01).Compared with model group,mangiferin significantly decreased serum levels of IL-6,TNF-αand IL-4 in renal tissues(P<0.05 or P<0.01).Compared with the model group,the levels of serum IL-10 and IFN-γin renal tissues were significantly increased in the middle and high doses mangiferin groups(P<0.05 or P<0.01).Conclusion The results showed that hypertension was associated with inflammatory reaction,and mangiferin had obvious anti-inflammatory effects.The mechanism may be related to the regulation of the proportion of T lymphocyte subsets,reducing the production of proinflammatory factors and the symptoms of hypertension and the damage of target organs.
